Transcriptional profile of Mouse liver after MitoCDNB application

ABSTRACT: To assess the effect of MitoCDNB on transcriptional profile of mouse liver tissue we injected mice with either vehicle (0.1 % ethanol) or 5 mg/kg of MitoCDNB. Liver was taken from both vehicle or MitoCDNB injected mice after 1 or 4 hours and the transcriptome assessed using Illumina RNASeq.

Methods: Adult male mice were exposed to Auditory Fear Conditioning training and subsequently injected with 3.0 mg/kg corticosterone, 20 mg/kg CORT108297, 80 mg/kg CORT118335, 40 mg/kg RU486 or vehicle. Dorsal hippocampi were dissected 3 hours after injection and snap-frozen. Total RNA was isolated and send for 100bp paired-end sequencing by BGI.

Transcriptome analysis of garlic induced hepatoprotection against alcoholic fatty liver

Project description:Alcohol induced fatty liver cause a dangerous health problem and is the major cause of morbidity and mortality worldwide. Garlic (Allium sativum) is documented to possess anti-fatty liver properties. However the exact molecular mechanisms are unknown. The main aim of this experiment is to elucidate the underlying pathways through which garlic ameliorates alcohol induced fatty liver. Dially disulfide and garlic oil were the garlic compounds used in this study. Leiber DeCarli ethanol liquid diet was to induce fatty liver in C57BL/6 mice model. Also the expression impaired by alcohol induced fatty liver is another aim of this study. Leiber-Decarli ethanol diet was used to induce fatty liver in male C57BL/6 mice (n=12). For control, Lieber-DeCarli liquid control diet was fed to mice (n=4). The control mice were pair-fed to the ethanol mice. After adaptation, the ethanol fed mice were divided into three groups viz. alcohol (n=4), dially disulfide [DADS] (n=4) and garlic oil [GO] (n=4). The study started with the administration of DADS (15 mg/kg bw) or GO (50 mg/kg bw) mixed in 0.1 ml olive oil through gavage. For the control and alcohol groups, same amount of olive oil (0.1 ml) was gavaged. The mice were gavaged daily for 4 weeks. The mice were euthanized by CO2 and blood was collected by cardiac puncture. Liver, kidney, spleen, lungs and hearts were collected and their weights recorded. A portion of liver was snap frozen in liquid nitrogen (200 mg) for RNA extraction.

Transcriptomic analysis of mouse subcutaneous tumor after intratumoral treatment with SLR14 or vehicle

Project description:We subcutaneously injected 5x105 YUMMER1.7 melanoma into right flank of C57BL/6J mice. 7 days later, the mice were intratumorally treated with 1 mg/kg SLR14 or vehicle every 3 days, for a total of 3 cycles. We observed a significant tumor growth delay in SLR14-treated mice compared to vehicle-treated mice.

Project description:Males were placed onto an all liquid diet with gradually increasing amounts of ethanol as follows: no ethanol for 2 days, 1% (v/v) for 2 days, 2% for 2 days, 4% for 1 week, 5% for 1 week, and 6% for 3 weeks. At the end of the experiment, one group from each genotype was injected with 5 mg/kg LPS. Mice were then euthanized 24 hours later.
