Project description:Smallpox has played an unparalleled role in human history and remains a significant potential threat to public health. Despite the historical significance of this disease, we know little about the underlying pathophysiology or the virulence mechanisms of the causative agent, variola virus. To improve our understanding of variola pathogenesis and variola-host interactions, we examined the molecular and cellular features of hemorrhagic smallpox in cynomolgus macaques. We used cDNA microarrays to analyze host gene expression patterns in sequential blood samples from each of 22 infected animals. Variola infection elicited striking and temporally coordinated patterns of gene expression in peripheral blood. Of particular interest were features that appear to represent an IFN response, cell proliferation, immunoglobulin gene expression, viral dose-dependent gene expression patterns, and viral modulation of the host immune response. The virtual absence of a tumor necrosis factor /NF-B-activated transcriptional program in the face of an overwhelming systemic infection suggests that variola gene products may ablate this response. These results provide a detailed picture of the host transcriptional response during smallpox infection, and may help guide the development of diagnostic, therapeutic, and prophylactic strategies.

Project description:The global spread of Monkeypox virus (MPXV) has resulted in the urgent need for an in-depth molecular characterization of the virus infection. Multiple omics studies on Vaccinia virus have extended our knowledge of poxvirus pathophysiology (REFs). Nevertheless, there are no comparative studies that would include an in-depth comparison of MPXV with other poxviruses in the context of molecular biology. Here, we report a comparative time-resolved proteomic study of MPXV and vaccinia viruses and a concurrent multi-omics study of MPXV infection in primary human cells. Using state-of-the-art proteomics, we profiled the virus-host interplay on the transcriptome, proteome and phosphoproteome levels in primary human foreskin fibroblasts. Pathway analysis in combination with projecting the gathered data onto the global network of host-signaling interactions revealed crosstalk between the perturbations at different levels, enabling identification of distinct and common molecular mechanisms of poxviruses. The NF-κB pathway, a signaling pathway governing immunomodulation and inflammation, was unexpectedly activated by MPXV but not VacV, while MPXV exhibited an astounding resistance to the effects of antiviral interferon response, potentially explaining the unique pathogenicity hallmarks of the monkeypox. Our extensive dataset highlights many signaling events and hotspots that influence the MPXV life cycle and may be used to guide rational design of both virus- and host-directed therapies, which we exemplify by identifying inhibitors of XXX, YYY, and ZZZ with potent antiviral efficacy against MPXV and/or VacV.

Project description:Evolutionarily successful poxviruses presented effective and diverse strategies to circumvent or overcome host defense mechanisms. Poxviruses encode many immunoregulatory proteins to evade host immunity for a productive infection and unique means of inhibiting DNA-sensing dependent type 1 interferon (IFN-I) responses is anticipated due to the biology of its dsDNA genome in nature and an exclusive cytoplasmic life cycle. We found the key DNA sensing inhibition by poxvirus infection was dominant during the early stage of poxvirus infection independent from DNA replication. In an effort of identifying poxvirus novel means to subdue antiviral proinflammatory responses e.g., IFN-I response, we focused on the function of one early gene that is the known host range determinant from the highly conserved poxvirus host range C7L superfamily, myxoma virus (MYXV) M062. Host range factors are unique features of poxviruses that determine the species and cell type tropism. Almost all sequenced mammalian poxviruses retain at least one homologue of the poxvirus host range C7L superfamily. In MYXV, a rabbit specific poxvirus, the dominant and broad-spectrum host range determinant of the C7L superfamily is the M062R gene. M062R gene product is essential for MYXV infection in almost all cells tested from different mammalian species and specifically inhibits the function of host Sterile α Motif Domain-containing 9 (SAMD9), as M062R-null (ΔM062R) MYXV causes abortive infection in a SAMD9-dependend manner. In this study we investigated the immunostimulatory property of the ΔM062R. We found that the replication-defective ΔM062R infection activated host DNA sensing pathway in the cGAS dependent fashion and knocking down SAMD9 expression attenuated proinflammatory responses. Moreover, transcriptomic analyses showed a unique feature of host gene expression landscape that is different from dsDNA stimulated inflammatory state. This study built a link between the anti-neoplastic SAMD9 and the regulation of the innate immune responses.

Project description:Modified Vaccinia Ankara (MVA) was recently approved as a Smallpox vaccine. Transmission of Variola is by respiratory droplets, and MVA delivered by skin scarification (ss) protected mice far more effectively against lethal respiratory challenge with VACV than any other route of delivery, and at much lower doses. Comparisons of ss with intradermal, subcutaneous or intramuscular routes showed that MVAOVA ss-generated T cells were both more abundant and transcriptionally distinct. MVAOVA ss produced greater numbers of lung Ova-specific CD8+ TRM and was superior in protecting mice against lethal VACVOVA respiratory challenge. Nearly as many lung TRM were generated with MVAOVA ss compared to direct pulmonary immunization with MVAOVA, and both routes vaccination protected mice against lethal pulmonary challenge with VACVOVA. Strikingly, MVAOVA ss-generated effector T cells exhibited overlapping gene transcriptional profiles to those generated via direct pulmonary immunization. Overall, our data suggest that heterologous MVA vectors delivered via ss are uniquely well-suited as vaccine vectors for respiratory pathogens like COVID-19. In addition, MVA delivered via ss could represent a more effective dose-sparing smallpox vaccine.

Project description:To investigate the mechanisms used by memory CD4+ T cells to protect the skin from poxvirus infection, we infected LCMV immune mice with VacV on the right ear skin and VacV expressing the dominant MHC-II restricted epitope from LCMV (VacV-Ii-GP61) on the left ear skin. On day 3 after the co-infection, RNA from whole skin was isolated and gene expression was analyzed using RNA sequencing.
