RNAseq measurement of cortisol induced gene expression change under knockout of glucocorticoid receptor (nr3c1)

ABSTRACT: We found that GR mediates most of the effects of the treatment, and paradoxically, that the transcriptome of cortisol-treated larvae is more like that of larvae lacking a GR than that of larvae with a GR, suggesting that the cortisol-treated larvae develop GR resistance.

Project description:We found that a frameshift deletion introduced into exon 1 of zebrafish klf9 eliminates the DNA binding domain and significantly reduces expression of the mature transcript. RNA-seq shows that klf9 is required for the pro-inflammatory transcriptomic effects of chronic cortisol treatment. klf9 is one of a core set of genes consistently found to be upregulated by chronic cortisol exposure.

Response to low-dose cortisol treatment in zebrafish embryos [larvae]

Project description:Chronic early life stress increases adult susceptibility to numerous health problems linked to chronic inflammation. One way that this may occur is via glucocorticoid-induced developmental programming. To gain insight into such programming we treated zebrafish embryos with 1 micromolar cortisol and examined the effects on larvae. Treated larvae had elevated whole-body cortisol and glucocorticoid signaling, and up-regulated genes associated with defense response and immune system processes.

Hepatic transcriptome response to glucocorticoid receptor activation in rainbow trout.

Project description:Stress induced elevation in plasma cortisol levels is shown to be involved in the metabolic re-adjustments to stress but the genetic basis of this response is still unclear. Cortisol signaling in teleosts is thought to be mediated predominantly by the glucocorticoid receptor (GR), a ligand-bound transcription factor and previous studies have shown that RU486, a GR antagonist offsets corticosteroid signaling in teleosts. In order to elucidate the genetic basis of GR-mediated metabolic re-adjustments, we exposed in vitro primary culture of trout hepatocytes to cortisol (100 ng/ml; stressed levels) or RU486 (1000 ng/ml) or a combination of both (RU486 + Cortisol), and analyzed the transcriptional responses using a low density custom-made targeted trout cDNA array. These results were further validated using quantitative real-time PCR. Cortisol treatment significantly increased glucose production in hepatocytes and this response is blocked by RU486 suggesting the activation of GR-mediated corticosteroid signaling. Cortisol treatment significantly modulated the transcript abundance of several genes, previously shown to be involved in molecular and biochemical responses to stress and most of these effects are abolished by RU486, suggesting GR-mediated effects. In addition, cortisol treatment affected key genes involved in reproduction supporting previous findings on inhibitory effects of cortisol on reproduction. Taken together, our results provide evidence on the transcriptional changes involved in cortisol signaling in response to stress. Transcription profiling of human HeLa cells treated by various doses of cortisol

Project description:Glucocorticoids act through the glucocorticoid receptor (GR), a ligand activated transcription factor, to modulate gene expression profiles in target cells and tissues. The principal physiological glucocorticoid in humans, cortisol, is released in a pulsatile fashion from the adrenal gland, which results in oscillations in blood concentrations, with a period of 1 - 2 hours. The biological consequence of fluctuating cortisol concentrations has not been explored. To identify the role of cortisol concentration oscillations for target cell response a flow-through cell culture system was developed. Transcription profiling was performed in the absence and presence of cortisol. Cells were exposed to constant cortisol levels of 100ng/ml or 200 ng/ml or oscillating cortisol concentrations.

Cortisol acting through the glucocorticoid receptor is not responsible for exercise-enhanced growth but does affect the white skeletal muscle transcriptome in zebrafish (Danio rerio)

Project description:Forced sustained swimming exercise at optimal speed enhances growth in many fish species, particularly through hypertrophy of the white skeletal muscle. The exact mechanism of this effect has not been resolved yet. To explore the mechanism, we first subjected wild-type zebrafish to an exercise protocol validated for exercise-enhanced growth, and showed that exercised zebrafish, which indeed showed enhanced growth, had higher cortisol levels than the non-exercised controls. A central role was therefore hypothesized for the steroid hormone cortisol acting through the Glucocorticoid receptor (Gr). Second, we subjected wild-type zebrafish and zebrafish with a mutant Gr to exercise at optimal, suboptimal and super-optimal speeds and compared them with non-exercised controls. Exercised zebrafish showed growth enhancement at all speeds, with highest growth at optimal speeds. In the Gr mutant fish, exercise resulted in growth enhancement similar to wild-type zebrafish, indicating that cortisol cannot be considered as a main determinant of exercise-enhanced growth. Finally, the transcriptome of white skeletal muscle tissue was analysed by RNA sequencing. The results of this analysis showed that in the muscle tissue of Gr mutant fish a lower number of genes is regulated by exercise than in wild-type fish (183 versus 351). A cluster of 36 genes was regulated by exercise in both wild-type and mutant fish. In this cluster, genes involved in transcriptional regulation and protein ubiquitination were overrepresented. Since growth was enhanced similarly in both wild-type fish and mutants, these processes may play an important role in exercise-enhanced growth.

Microarray analysis of transcripts in the liver of gilthead sea bream with high plasma cortisol levels induced by slow-release cortisol implants

Project description:The present work was designed to assess the effects of high plasma cortisol levels induced by slow-release cortisol implants in the mRNA transcription of the GR in the different organs of the Sparus aurata, including liver. For that purpose fish were intraperitoneally injected with the implants containing two different concentrations of cortisol (50 or 200 µg/g body weight) and blood and organs were sampled after 7 and 14 days of implantation. Only fish with 200 µg/g implants exhibited a significant rise in the plasma cortisol. For microarray analysis we used livers of gilthead sea bream (N=36 fish). Fish were injected with 200 µg/g body weight of cortisol and sampled after 7 and 14 days (n=6 for each condition). RNA samples were grouped into pools of 2 animals for each time point. The analysis were carried out considering the transcripts differentially expressed in the group of fish implanted during 7 days with cortisol comparing to control group. Additionally, the transcripts differentially expressed at day 14 were compared with day 7.
