Project description:Tissue stem cells are required for homeostasis and wound repair throughout the entire lifespan. Multiple mechanisms have been extensively studied for the maintenance of stem cell quiescence. Here we use Omini-ATACseq and single cell ATACseq to study the stem cell quiescence at epigenetic level. We sought out to compare the open chromatin state in different cell lineages between control and Foxc1/Nfatc1 double knockout mice. The single cell ATACseq could further help infer the transcription regulation between enhancers and promoters interaction.

Project description:Hair follicle stem cells(HFSCs) can transition between active and quiescent stages during normal hair cycle or would healing. Transcriptional regulation has been show to regulate the transition. Previous studies have shown that two transcription factor, Foxc1 and Nfatc1, can help maintain the stem cells in quiescent state, the deletion of either one could lead to precocious activation. Here, we collect the hair follicle stem cell at postnatal day 30 from both control mice, Nfatc1 knockout mice, Foxc1 knockout mice(downloaded) and Foxc1/Nfatc1 double knockout mice to study the transcriptional regulation network of HFSCs quiescence.

Project description:Quiescent stem cells are periodically activated to maintain tissue homeostasis or occasionally called into action upon injury. Molecular mechanisms that constitutively maintain stem cell identity or promote stem cell proliferation and differentiation upon activation have been extensively studied. However, it is unclear how quiescent stem cells maintain identity and reinforce quiescence when they transition from quiescence to activation. Here we show mouse hair follicle stem cell compartment induces a transcription factor, Foxc1, when activated. Importantly, deletion of Foxc1 in the activated but not quiescent stem cells compromises stem cell identity, fails to re-establish quiescence and subsequently drives premature stem cell activation.These findings uncover a dynamic, cell-intrinsic mechanism employed by hair follicle stem cells to reinforce stemness in response to activation. Project description:We report downstream gene expression changes in stem cells of the adult mouse hair follicle upon conditional ablating of the transcription factor Forkhead Box C1 transcription factor (FOXC1). Hair follicles undergo cycles of rest (telogen; Tel) and regeneration (anagen; Ana). As such, we performed our analysis on these two different stages of hair follicles. mRNA-sequencing of WT vs. Foxc1-conditional or inducible KO (Foxc1-cKO or iKO) hair follicle stem cells (HFSCs) purified from mouse dorsal back skin by flow-activated cell sorting (FACS).

Project description:Aging organs functionally and structurally decline with the loss of their regenerative capabilities, yet the existence of distinct cell division programs that determine organ fates is unknown. Hair follicles, mammalian mini-organs that grow hair, miniaturize by aging. Here we report that hair follicle regeneration and aging are driven by distinct cell division types of hair follicle stem cells (HFSCs). Cell fate tracing and cell division axis analysis in mice revealed that HFSCs undergo symmetric and asymmetric cell divisions to generate a new bulge, yet preferentially provoke “stress-responsive type” asymmetric cell divisions that generate aberrantly differentiating cells upon age/stress. That dynamic program with repetitive divisions efficiently eradicates those cells through defective association and stabilization of a hemidesmosomal protein COL17A1 and a cell-polarity-protein aPKCλ in HFSCs, thereby causing organ aging. The forced stabilization of COL17A1 rescued organ aging through aPKCλ stabilization. These results demonstrate that distinct cell division programs govern tissue/organ fates.

Project description:Adult stem cells (SCs) are essential for tissue maintenance and regeneration yet are susceptible to SC senescence during aging. Here we demonstrate the importance of cellular NAD+ level and its impact on mitochondrial activity as a pivotal switch to modulate muscle stem cell (MuSC) senescence. Importantly, the induction of the mitochondrial unfolded protein response (UPRmt) and of prohibitin proteins, subsequent to increasing cellular NAD+ with the precursor nicotinamide riboside (NR), rejuvenates MuSCs in aged mice. NR also prevents MuSCs senescence in Mdx mice, a mouse model of muscular dystrophy. Extending these observations to other SC pools and on the organism as a whole, we demonstrate that NR delays neural stem cell (NSC) and melanocyte stem cell (McSC) senescence, while also increasing mouse lifespan. Strategies that conserve cellular NAD+ could therefore be utilized to reprogram dysfunctional SCs in aging and disease to improve lifespan in mammals
