Transcription profile of K-ras activated murine menstruating uteri

ABSTRACT: The expression of K-ras transgene is induced in K-rasV12+/-/Cre+/+/ROSA26R-LacZ+/+ transgenic mice uteri, which were primed with hormone treatment. Decidualization was artificially induced in these uteri and hormone injection was withdrawn to induced menstruation-like endometrial degeneration. Microarray was then used to study the molecular changes in these menstruating uteri following the K-ras activation.

Uterine tissues were collected from K-ras induced and littermate controled menstruating uteri. Experiments were carried out using Illumina Mouse WG-6 BeadChips (n=6 in each group). Data were normalized in R environment using the Lumi Bioconductor package.

Gene expression profile of peri-implantation uteri treated with LPA3 receptor agonist

Project description:To investigate gene expression changes induced by Lysophosphatidic acid receptor 3 (LPA3) activation in peri-implantation uteri, we performed microarray analysis in LPA3 agonist (T13)-injected pseudopregnant uteri. On 3.5 dpc, pseudopregnant mice were subjected with intrautero-injection of vehicle (0.01% BSA-PBS) or T13 and then uteri were dissected 24-36 hr after the injection. We found that several thousand genes were up- or down-regulated by T13 injection. Interestingly, well-known decidual markers such as Bmp2 and Wnt4 were dramatically induced by T13, which we certificated in quantitive RT-PCR.

Project description:The goal of this study is to profile gene expression patterns of RasV12-induced brain tumors and tumor-associated macrophages (TAM) in the cerebellum. Ras activation in Msi1+ cells induced tumor formation in the cerebellum of Msi1-CreER;RasV12 mice (Ras). GFP+ tumor cells and CD11b+ phagocytes were isolated from the cerebellum of Ras mice at 3 weeks and used for gene expression analysis. GFP+ cells and CD11b+ phagocytes isolated from the cerebellum of Msi1-CreER;GFP mice (Cont) were used as control.

Microarray analysis identified differentially expressed genes in the glandless uteri of PUGKO mice as compared to control mice.

Murine model of decidulization and menstruation

Project description:A murine model that mimic the decidualization and regression observed in human was used to investigate the molecular mechanisms underlying the dynamic processes in endometrium. Ovariectomized mice were treated sequentially with steroid hormones and then, to induce decidualization, oil was injected into the uterine lumen. A process similar to menstruation was induced by hormone-withdrawal. The uterine tissues were collected at 4 time-points after the induction of decidualization. Transcription profiling of mouse uteri after ovariectomy and DHT treatment.

Project description:Morphogenesis of epithelial tissues relies on the precise developmental control of cell polarity and architecture. In the early Drosophila embryo, the primary epithelium forms during cellularisation, following a tightly controlled genetic programme where specific sets of genes are up-regulated. Some of them, for instance, control membrane invagination between the nuclei anchored at the apical surface of the syncytium. We study the whole genome expression profile aftre DHT treatment in the uteri of gonadectomized mouse. Experiment Overall Design: Ten 10-11 weeks old C57Bl/6 mice were gonadectomized under isoflurane-induced anesthesia and sacrified seven days after GDX. Mice were injected s.c with DHT (0.1mg/mice) or vehicle, at 0.5, 1, 3, 6, 12 or 24 h before sacrifice. Uteri was removed and pooled before total RNA extraction. Samples was hybridized to U74Av2 Genechips (Affymetrix).
