Project description:The fetal inflammatory response (FIR) increases the risk of perinatal brain injury, particularly in extremely low gestational age newborns (ELGANs, < 28 weeks of gestation). One of the mechanisms contributing to such a risk is a postnatal intermittent or sustained systemic inflammation (ISSI) following FIR. The link between prenatal and postnatal systemic inflammation is supported by the presence of well-established inflammatory biomarkers in the umbilical cord and peripheral blood. However, the extent of molecular changes contributing to this association is unknown. Using RNA sequencing and mass spectrometry proteomics, we profiled the transcriptome and proteome of archived neonatal dried blood spot (DBS) specimens from 21 ELGANs. Comparing FIR-affected and unaffected ELGANs, we identified 782 gene and 27 protein expression changes of 50% magnitude or more, and an experiment-wide significance level below 5% false discovery rate. These expression changes confirm the robust postnatal activation of the innate immune system in FIR-affected ELGANs and reveal for the first time an impairment of their adaptive immunity. In turn, the altered pathway provide clues about the molecular mechanisms triggering ISSI after FIR, and the onset of perinatal brain injury.

Project description:<p><strong>INTRODUCTION:</strong> Restricted or enhanced intrauterine growth is associated with elevated risks of early and late metabolic problems in humans. Metabolomics based on amino acid and carnitine/acylcarnitine profile may have a role in fetal and early postnatal energy metabolism. In this study, the relationship between intrauterine growth status and early metabolomics profile was evaluated.</p><p><strong>MATERIALS AND METHODS:</strong> A single center retrospective cohort study was conducted. Newborn infants were enrolled into the study and they were grouped according to their birth weight percentile as small for gestational age (SGA), appropriate for gestational age (AGA) and large for gestational age (LGA) infants. In all infants amino acid and carnitine/acylcarnitine profile with liquid chromatography-tandem mass spectrometry (LC-MS/MS) were recorded and compared between groups.</p><p><strong>RESULTS:</strong> LGA infants had higher levels of glutamic acid and lower levels of ornithine, alanine and glycine (p<0.05) when compared to AGA infants. SGA infants had higher levels of alanine and glycine levels when compared with AGA and LGA infants. Total carnitine, C0, C2, C4, C5, C10:1, C18:1, C18:2, C14-OH and C18:2-OH levels were significantly higher and C3 and C6-DC levels were lower in SGA infants (p<0.05). LGA infants had higher C3 and C5:1 level; and lower C18:2 and C16:1-OH levels (p<0.05). There were positive correlations between free carnitine and phenylalanine, arginine, methionine, alanine and glycine levels (p<0.05). Also a positive correlation between ponderal index and C3, C5-DC, C14, C14:1; and a negative correlation between ponderal index and ornithine, alanine, glycine, C16:1-OH, C18:2 were shown.</p><p><strong>CONCLUSION:</strong> We demonstrated differences in metabolomics possibly reflecting the energy metabolism in newborn infants with intrauterine growth problems in the early postnatal period. These differences might be the footprints of metabolic disturbances in future adulthood.</p>

Project description:The developing brain is particularly sensitive to ethanol during the brain growth spurt or synaptogenesis (third human trimester equivalent). This has been shown to lead to abnormal brain development and behavioural changes in the adult mouse that are relevant to those seen in humans with fetal alcohol spectrum disorders (FASD). We evaluated the acute (4h post-treatment) gene expression changes that occur in the brain due to ethanol exposure during synaptogenesis (postnatal day 7). We used microarray analyses to evaluate the changes in brain gene expression at postnatal day 7 that occur due to ethanol treatment at postnatal day 7 (synaptogenesis).

Project description:Development of the brain involves the formation and maturation of numerous synapses. This process requires prominent changes of the synaptic proteome and potentially involves thousands of different proteins at every synapse. To date the proteome analysis of synapse development has been studied sparsely. Here, we analyzed the cortical synaptic membrane proteome of juvenile [postnatal day 9 (P9), P15, P21, P27], adolescent (P35) and different adult ages (P70, P140, P280) of C57Bl6/J mice. Using a quantitative proteomics workflow we quantified 1560 proteins of which 696 showed statistically significant differences over time. Proteins of the presynaptic active-zone and postsynaptic element showed increased levels over time; proteins involved in protein synthesis or neurite outgrowth generally decreased in abundance. In several cases, proteins from a single functional molecular entity, e.g., subunits of the NMDA receptor, showed profound differences in their temporal regulation, which may reflect specific synaptic development features of connectivity, strength and plasticity.

Project description:Background: NRF2 is an essential cytoprotective transcription factor inducing antioxidant response element (ARE)-bearing genes. However, association of NRF2 with lung development has not been examined. Human lungs are not fully developed until 2-3 years of and they are fully matured at about 8 years. Murine lungs at birth are immature (at saccular stage of lung development) and have been used to study developmental lung disorders. Methods: To investigate (1) the transcriptome changes during lung development and (2) the role of NRF2 in lung development and maturation in mice, lungs were harvested from Nrf2-deficient (Nrf2-/-) and wild-type (Nrf2+/+) mouse embryos, neonates and adults. Microarray and pathway analysis determined NRF2-directed mechanisms underlying lung development and maturation. Results: Nrf2 mRNA expression was peack at embryonic days E17.5-E18.5 (immediately before birth) probably to increase antioxidant apparatus to prepare against high O2 environment after birth. The pseudoglandular phase lungs (E13-E15) are undergoing vigorous cell proliferation under the control of high-fidelity DNA damage repair system. Fetal lungs (E13.5-E17.5) are lack in immune system, xenobiotic metabolism, and tissue damage genes. After birth at postnatal day 1 (PND1), lung cell division is quiescent but transporters and lipid metabolism are activated. When lung enters alveolar phase (PND4), cell proliferation is resumed. Mature lungs (PND14-P42) have heightened networks of host defense systems (immunity, antioxidants) and cellular injury and abnormality (e.g., glucose metabolism disorder). Nrf2 deletion in fetal lung (E13.5-E17.5) altered developmental, immunity, and metabolism genes, and it may have affected lung branching. Nrf2 deletion affected lung transcriptome changes the most at E17.5 when Nrf2 message level is maximum (E17.5-E18.5). Nrf2 deletion in newborn lung (PND0) decreased cell cycle progress and DNA damage repair. Nrf2 deletion in neonatal lung (PND1-4) enhanced tissue injury/cell death and inhibited developmental cell differentiation. Nrf2 deletion in matured mouse lung (PND42) affected not only antioxidant pathway but also immune responses and connective tissue cell migrations. Conclusion: Overall, NRF2 plays multiple roles in underdeveloped lungs and associated with lung morphogenesis, immunity, cell cycle progress, tissue differentiation and metabolism as well as cellular defense. Results provide putative molecular mechanisms of NRF2-directed lung morphogenesis and maturation.

Project description:Pregnancy is a time of extreme metabolic demand that requires coordinated adaptations between mother and fetus. To determine the contributions of maternal and fetal metabolism to metabolic plasticity during gestation, mice with a liver-specific Carnitine Palmitoyltransferase-2 knockout mice (Cpt2-/-), or Pparα KO mice were subjected to late-gestation nutrient stress, a 24hr fast from E16.5 to E17.5. The fetal response to maternal fasting was dominated by maternal lipid metabolism as the loss of maternal hepatic fatty acid oxidation or Pparα signaling accelerated fetal liver transcriptional programing. These data show that maternal nutritional environment is a major driver of perinatal metabolic programing and plasticity.
