Project description:Regulation of stomatal movement is one of the effective strategies for developing resistant crops to air pollutant because stomata allows absorption of various air pollutants, such as ozone and sulfur dioxide. Transcription factor (TF) is a fascinating target of genetic manipulation for this end because TF regulates many genes simultaneously and methods of genetic manipulation are universally established. Here, we have screened transgenic Arabidopsis lines expressing chimeric repressor of TFs in high-concentration of ozone and found that the chimeric repressors of GOLDEN-LIKE1 (GLK1) and GLK2 (GLK1-SRDX and GLK2-SRDX) conferred strong tolerance to ozone. These 35S:GLK1/2-SRDX plants also showed sulfur dioxide tolerance. Their leaves showed lower rate of transpiration than wild type and a remarkable closed-stomata phenotype. The expression of the genes encoding K+ and water channels, including KAT1 and AKT1, which are involved in stomatal opening, was downregulated in 35S:GLK1-SRDX plants. Consistently, expression of GLK1-SRDX driven by the GC1 promoter, which has an activity only in guard cell, also induced closed-stomata and an ozone tolerant phenotype. On the contrary, 35S:GLK1/2 plants showed hypersensitivity to ozone and an opened-stomata phenotype. These data suggested that GLK1 and GLK2 have an ability to induce transcriptional change in guard cell and regulate stomatal movement. Our findings provide an effective tool to confer resistance to air pollutant by regulating stomatal aperture and improve crop productivity in future.

Project description:Relatively few studies have evaluated the effects of heterogeneous spatiotemporal pollutant distributions on health risk estimates in time-series analyses that use data from a central monitor to assign exposures. We present a method for examining the effects of exposure measurement error relating to spatiotemporal variability in ambient air pollutant concentrations on air pollution health risk estimates in a daily time-series analysis of emergency department visits in Atlanta, Georgia. We used Poisson generalized linear models to estimate associations between current-day pollutant concentrations and circulatory emergency department visits for the 1998-2004 time period. Data from monitoring sites located in different geographical regions of the study area and at different distances from several urban geographical subpopulations served as alternative measures of exposure. We observed associations for spatially heterogeneous pollutants (CO and NO(2)) using data from several different urban monitoring sites. These associations were not observed when using data from the most rural site, located 38 miles from the city center. In contrast, associations for spatially homogeneous pollutants (O(3) and PM(2.5)) were similar, regardless of the monitoring site location. We found that monitoring site location and the distance of a monitoring site to a population of interest did not meaningfully affect estimated associations for any pollutant when using data from urban sites located within 20 miles from the population center under study. However, for CO and NO(2), these factors were important when using data from rural sites located > or = 30 miles from the population center, most likely owing to exposure measurement error. Overall, our findings lend support to the use of pollutant data from urban central sites to assess population exposures within geographically dispersed study populations in Atlanta and similar cities.

Project description:Short title: Coral Meta-Transcriptomics Reveal Pollutant Stress Background: Corals represent symbiotic meta-organisms that require harmonization among the coral animal, photosynthetic zooxanthellae and associated microbes to survive environmental stresses. We investigated integrated-responses among coral and zooxanthellae in the scleractinian coral Acropora formosa in response to an emerging marine pollutant, the munitions constituent, 1,3,5-trinitro-1,3,5 triazine (RDX; 5 day exposures to 0 (control), 0.5, 0.9, 1.8, 3.7, and 7.2 mg/L, measured in seawater). Results: RDX accumulated readily in coral soft tissues with bioconcentration factors ranging from 1.1 to 1.5). Next-generation sequencing of a normalized meta-transcriptomic library developed for the eukaryotic components of the A. formosa coral holobiont was leveraged to conduct microarray-based global transcript expression analysis of integrated coral / zooxanthellae responses to the RDX exposure. Total differentially expressed transcripts (DET) increased with increasing RDX exposure concentrations as did the proportion of zooxanthellae DET relative to the coral animal. Transcriptional responses in the coral demonstrated higher sensitivity to RDX compared to zooxanthellae where increased expression of gene transcripts coding xenobiotic detoxification mechanisms (ie. cytochrome P450 and UDP glucuronosyltransferase 2) were initiated at the lowest exposure concentration. Increased expression of these detoxification mechanisms was sustained at higher RDX concentrations as well as production of a physical barrier to exposure through a 40% increase in mucocyte density at the maximum RDX exposure. At and above the 1.8 mg/L exposure concentration, DET coding for genes involved in central energy metabolism, including photosynthesis, glycolysis and electron-transport functions, were decreased in zooxanthellae although preliminary data indicated that zooxanthellae densities were not affected. In contrast, significantly increased transcript expression for genes involved in cellular energy production including glycolysis and electron-transport pathways was observed in the coral animal. Conclusions: Transcriptional network analysis for central energy metabolism demonstrated highly correlated responses to RDX among the coral animal and zooxanthellae indicative of potential compensatory responses to lost photosynthetic potential within the holobiont. These observations underscore the potential for complex integrated responses to RDX exposure among species comprising the coral holobiont and highlight the need to understand holobiont-species interactions to accurately assess pollutant impacts.

Project description:Composting can divert organic waste from landfills, reduce landfill methane emissions, and recycle nutrients back to soils. However, the composting process is also a source of greenhouse gas and air pollutant emissions. Researchers, regulators, and policy decision-makers all rely on emissions estimates to develop local emissions inventories and weigh competing waste diversion options, yet reported emission factors are difficult to interpret and highly variable. This review explores the impacts of waste characteristics, pretreatment processes, and composting conditions on CO2, CH4, N2O, NH3, and VOC emissions by critically reviewing and analyzing 388 emission factors from 46 studies. The values reported to date suggest that CH4 is the single largest contributor to 100-year global warming potential (GWP100) for yard waste composting, comprising approximately 80% of the total GWP100. For nitrogen-rich wastes including manure, mixed municipal organic waste, and wastewater treatment sludge, N2O is the largest contributor to GWP100, accounting for half to as much as 90% of the total GWP100. If waste is anaerobically digested prior to composting, N2O, NH3, and VOC emissions tend to decrease relative to composting the untreated waste. Effective pile management and aeration are key to minimizing CH4 emissions. However, forced aeration can increase NH3 emissions in some cases.

Project description:IntroductionThe developing fetus relies on the maternal blood supply to provide the choline it requires for making membrane lipids, synthesizing acetylcholine, and performing important methylation reactions. It is vital, therefore, that the placenta is efficient at transporting choline from the maternal to the fetal circulation. Although choline transporters have been found in term placenta samples, little is known about what cell types express specific choline transporters and how expression of the transporters may change over gestation. The objective of this study was to characterize choline transporter expression levels and localization in the human placenta throughout placental development.MethodsWe analyzed CTL1 and -2 expression over gestation in human placental biopsies from 6 to 40 weeks gestation (n = 6-10 per gestational window) by immunoblot analysis. To determine the cellular expression pattern of the choline transporters throughout gestation, immunofluorescence analysis was then performed.ResultsBoth CTL1 and CTL2 were expressed in the chorionic villi from 6 weeks gestation to term. Labor did not alter expression levels of either transporter. CTL1 localized to the syncytial trophoblasts and the endothelium of the fetal vasculature within the chorionic villous structure. CTL2 localized mainly to the stroma early in gestation and by the second trimester co-localized with CTL1 at the fetal vasculature.DiscussionThe differential expression pattern of CTL1 and CTL2 suggests that CTL1 is the key transporter involved in choline transport from maternal circulation and both transporters are likely involved in stromal and endothelial cell choline transport.
