Differential expression of Aspergillus genes during the induction of aflatoxin biosynthesis

Project description:Gene expression analysis of A. parasiticus grown under conditions conducive and nonconductive for aflatoxin production was evaluated using glass slide microarrays containing the 753 ESTs. A complex regulatory network governs the biosynthesis of aflatoxin. While several genes involved in aflatoxin production are known, their action alone cannot account for its regulation. Arrays of clones from an Aspergillus flavus cDNA library and glass slide microarrays of ESTs were screened to identify additional genes. An initial screen of the cDNA clone arrays lead to the identification of 753 unique ESTs. Many showed sequence similarity to known metabolic and regulatory genes; however, no function could be ascribed to over 50% of the ESTs. Gene expression analysis of Aspergillus parasiticus grown under conditions conducive and non-conductive for aflatoxin production was evaluated using glass slide microarrays containing the 753 ESTs. Twenty-four genes were more highly expressed during aflatoxin biosynthesis and 18 genes were more highly expressed prior to aflatoxin biosynthesis. No predicted function could be ascribed to 18 of the 24 genes whose elevated expression was associated with aflatoxin biosynthesis. Keywords = Aspergillus Keywords = aflatoxin Keywords: time-course

Transcription profiling of 23 day old Arabidopsis plants, wild type Col-0 and mutant rcd1

Project description:23 day old Arabidopsis plants, wildtype Col-0 and mutant rcd1, were compared to determine which genes are over- or under expressed in the mutant. Six biological repeats were pooled in pairs of two for labellings. All labellings/hybridizations were done with a dye swap technical repeat (e.g. in total six labellings were used). The Turku Arabidopsis 8K arrays were used for hybridizations. For the hybridizations the samples were split in two halves with each half hybridized to Turku Arabidopsis 8K slide 1 and Turku Arabidopsis 8K slide 2 (e.g. in total 12 hybridizations were performed).

Genomic profile of maize response to A. flavus infection identifies genes associated with biotic and abiotic resistance

Project description:The experiment consisted of a field design containing two maize inbred lines Va35 and Mp313E. Va35 has yellow kernels and is susceptible to kernel infection by Aspergillus flavus (A. flavus). Mp313E is a white dent maize inbred line and was released primarily as a source of resistance to kernel infection by A. flavus. The test ears were inoculated with Aspergillus flavus and collected two days after inoculation. Non-inoculated ears collected 16 days after pollination were also used as a control. The microarray experimental design was a randomized complete block design with three replications. One microarray slide was used for each field plot. Each slide contained the inoculated and non-inoculated sub-treatments. A second slide for one replication of each genotype contained a dye swap for the inoculation treatment and three subsamples (dots) within the slide represented each contig. Most of rice miRNAs have no significant change. miR-169f showed osmotic-responsive up-regulation. However, the others members of miR-169 family displays no responses to the osmotic stress. For each sample, two independent small RNA libraries (biological replications) were generated and two independent reverse transcription/amplifications/hybridizations (technical replications) were performed for each library. Thus, for each sample data was collected from four independent hybridizations.

Transcription profiling of Gerbera hybrida flower organs vs a reference pool

Project description:Different gerbera hybrida flower organs including pappus bristles, stamen, early and late petal development and flower scape, were compared to a pooled reference sample to determine which genes are abundant for analyzed flower organs. Samples were pooled and three replicants that were independently labelled were included. All labellings/hybridizations included one dye swap technical repat. Gerbera 9K cDNA microarrays were used for hybridizations.
