Project description:Olfactory sensory neurons (OSNs) expressing the same olfactory receptor (OR) are randomlydispersed within the same olfactory epithelial zone. To date, between 4 and 12 zones havebeen defined based on OR expression by in-situ hybridization studies. However, the totalnumber of zones, their organisation, molecular identity, and their distribution across thedorsal-ventral, anterior-posterior and lateral-medial axes are still unknown. To address thesequestions we propose to perform RNA-seq on sequential cryosections along the dorsalventraland anterior-posterior axes. This will provide a spatial dimension to the quantitativedata we already have on the number of each neuron type in the entire epithelium. We aim touse this as preliminary data for a grant application to completely resolve the 3D organisationof the olfactory epithelium at a cellular level. This data is part of a pre-publication release. Project description:Expression profiling of mRNA abundance in the adult mouse olfactory epithelium during replacement of OSNs forced by the bilateral ablation of the olfactory bulbs. The experiment was done on 6 week old male C57Bl/6 mice. Olfactory epithelium tissue samples were collected on days 1, 5, and 7 after bulbectomy. The cellular processes activated by bulbectomy include apoptosis of mature olfactory sensory neurons, infiltration of macrophages and dendritic cells, stimulation of proliferation of basal cell progenitors, and differentation of new sensory neurons.

Expression data from Emx2 wildtype and knockout olfactory epithelium

Project description:Emx2 is a homeobox transcription factor that plays a critical role in development. Olfactory sensory neuron axons from Emx2 knockout mice fail to innervate their target tissue, the olfactory bulb. Homeobox transcription factors may also play an important role in olfactory receptor expression. We used microarrays to analyze differences in mRNA abundance in the olfactory epithelium of Emx2 wildtype and knockout embryonic day 18.5 mice. Keywords: Knockout-Wildtype comparision Total RNA from 9 Emx2 wildtype and 9 Emx2 knockout mice was analyzed. Equal amounts of RNA from 3 animals was pooled to create one biological sample, 3 pools per genotype, 6 chips total. RNA was hybridized to the Affymetrix Mouse Exon 1.0 ST array.

Transcription profiling of rat nasal respiratory and olfactory epithelium

Project description:The nasal epithelium is an important target site for chemically-induced toxicity and carcinogenicity. Human epidemiological investigations and experimental laboratory animal studies show that the nasal olfactory epithelium is selectively damaged by inhalation exposure to several chemicals, including vinyl acetate and hydrogen sulfide. The reason for the relative sensitivity of the nasal olfactory epithelium is not known. To better understand and predict the response of the nasal epithelium to inhaled xenobiotics, gene expression profiles from naÑve male and female Sprague-Dawley rats were constructed. Epithelial cells were manually collected from the nasal septum, naso- and maxillo-turbinates, and ethmoid turbinates of 9 male and 9 female rats. Gene expression analysis was performed using the Affymetrix Rat Genome 430 2.0 microarray. Gene ontology enrichment analysis identified several functional categories including xenobiotic metabolism, cell cycle, apoptosis, and ion channel/transport with significantly different expression between tissue types. Surprisingly, there were few gender differences in gene expression. This baseline data will contribute to our understanding of the normal physiology and selectivity of the nasal epithelial cellsâ response to inhaled environmental toxicants. Experiment Overall Design: To better understand and predict the response of the nasal epithelium to inhaled xenobiotics, gene expression profiles from naÑve male and female Sprague-Dawley rats were constructed. Epithelial cells were manually collected from the nasal septum, naso- and maxillo-turbinates, and ethmoid turbinates of 9 male and 9 female rats. Gene expression analysis was performed using the Affymetrix Rat Genome 430 2.0 microarray.
