Meg-01 cell line expression data after propionate stimulation

ABSTRACT: Platelets are produced by megakaryocytes, deriving from megakaryocyte erythrocyte progenitors (MEP) in the bone marrow. Increased megakaryocyte expansion across common autoimmune diseases was shown for rheumatoid arthritis (RA), systemic lupus erythematosus (SLE) and primary Sjögren’s syndrome (pSS). In this context, we evaluated the role of the microbial-derived short chain fatty acid (SCFA) propionate on megakayriocytes in vitro. Using RNA sequencing we characterized the consequences of propionate exposure on the megakaryoblast cell line Meg-01.

Project description:GAS2DN could suppress the growth of chronic myeloid leukemia cells, including K562, MEG-01 and CD34+ cells from patients. In addition, GAS2DN inhibited the tumorigenic ability of MEG-01 cells in nude mice. To understand the molecular insight of this inhibitory effect of GAS2DN, global gene expression were performed. The control and GAS2DN-transduced MEG-01 cells were used for microarray analysis. Three biological independent extracts of control and GAS2DN-transduced cells were pooled together with equal amount, and then the pooled samples were compared with Affymetrix chips.

meg-3 meg-4 upregulate endogenous sRNAs targeting RNAi genes and downregulate their mRNAs

Project description:sRNAseq was conducted on four independently generated meg-3 meg-4 lines, and upregulation of sRNAs targeting several RNAi genes was found. mRNAseq found that the transcript levels for several of these genes was also down. meg-3 meg-4 mutants also showed a similar pattern of sRNA misregulation as the rde-11 mutant.

Transcription profiling by array of parotid tissue from Primary Sjögren’s Syndrome and controls

Project description:Primary Sjögren’s syndrome (pSS) is a chronic autoimmune disease with complex etiopathogenesis. Here we use Affymetrix U133 plus 2.0 microarray gene expression data from human parotid tissue. Parotid gland tissues were harvested from 17 pSS and 14 14 non-pSS sicca patients and 18 controls. The data were used in the following article: Nazmul-Hossain ANM, Pollard RPE, Kroese FGM, Vissink A, Kallenberg CGM, Spijkervet FKL, Bootsma H, Michie SA, Gorr SU, Peck AB, Cai C, Zhou H, Horvath S, Wong DTW (2012) Systems Analysis of Primary Sjögren’s Syndrome Pathogenesis in Salivary Glands: Comparative Pathways and Molecular Events in Humans and a Mouse Model. Gene expression data in Meg-01 cells following transfection with platelet-enriched microRNA-1225-3p

Project description:Platelets from sickle cell disease patients have differentially expressed microRNAs as compared to platelets from healthy control subjects. The objective of this study is to overexpress an upregulated miRNA,miR-1225-3p, in MEG01 cells to identify putative targets. MEG-01 cells were transfected with microRNA-1225-3p mimic for overexpression. Simultaneously, a negative control (scrambled) RNA was transfected for comparison. Gene expression was measured at 24 hours after transfection. Five independent experiments were performed at the same time in each group: microRNA-1225-3p transfected and scrambled.
