RNA sequencing of human peripheral blood cDC1 treated with PGE2

ABSTRACT: RNA sequencing was used to characterize PGE2-mediated changes in the gene expression profile of human conventional type 1 dendritic cells (cDC1) purified from PBMCs of healthy donors. Our analysis shows that treatment of cDC1 with PGE2 induces transcriptional changes in resting cDC1. cDC1 activated with a TLR3 ligand after PGE2 pre-treatment show alterations in the expression of activation induced genes.

PGE2 signaling networks in T cells

Project description:We combined phosphoproteomics and phosphoflow cytometry with specific stimulation of the four PGE2 receptors to obtain a global overview of PGE2-regulated signaling pathways and networks in T cells. The study revealed phosphoproteomes regulated uniquely by each receptor and crosstalk between receptors. Previously unknown PGE2-regulated phosphosites were identified, including biologically significant sites.

PGE2 effect on unstimulated and exhausted human CD8+ T-cells

Project description:Prostaglandin E2 (PGE2) is overexpressed in many tumors and plays an important role in suppressing T-cell function. In this study, we investigated the effect of PGE2 in human T-cells. Importantly, PGE2 caused profound transcriptomic changes both in unstimulated and exhausted T-cells with many genes and pathways altered including T-cell lipid metabolism. Our study suggests that PGE2-induced metabolic reprogramming is detrimental to dysfunctional T-cells and TIL, and its manipulation could be used to dampen the immunosuppressive effects of PGE2.
