Project description:Purpose: To clarify the function of TIM-3 signaling in AML cells. Analysis of lentiviral shRNA-mediated knockdown of TIM-3 in TIM-3+ acute myeloid leukemia (AML) cell line, KASUMI-3. Consistent with our previous study (Kikushige et al., Cell Stem Cell 2015) revealing the presence of TIM-3/galectin-9 autocrine loop and its constitutive signaling in AML, KD of receptor,TIM-3, alone significantly impaired the proliferation of KASUMI-3 in vitro and in vivo.

Project description:To investigate the function of p53 in regulating response to MET inhibition in MPNST cells we established NF1-MET;sgP53 cells in which Trp53 was knocked out using CRISPR-Cas9. As a control, the parental NF1-MET was transduced with CRISPR-Cas9 in the absence of sgRNA. We then performed gene expression profiling analysis using data obtained from RNA-seq of the 2 different cells treated with either capmatinib or vehicle in duplicate.

Effect of Rgnef knockout in an aggressive ovarian cancer cell line

Project description:CRISPR/Cas9-mediated Rgnef knockout was performed in the aggressive ID8-IPs (alternaate name is KMF cells). Gene expression profiles of Rgnef-/- or Rgnef-/- cells re-expressing GFP-Rgnef were generated. Differential downregulation of antioxidant genes was observed in Rgnef-/- cells. Results provide insight into the role of Rgnef in promoting ovarian tumor progression.
