Effect of exosomal miRNA of cancer-associated adipocyte (CAA) on the malignant transformation of pancreatic ductal adenocarcinoma

ABSTRACT: CAAs were generated by co-culturing human adipocytes with a human pancreatic cancer cell line (Panc-1), and exosomes were isolated from the CAA-conditioned medium (CAA-CM). We then performed gene expression profiling analysis using small RNA-seq of three differernt CAA-CMs and normal A-CMs.

Inhibition of PPARg by BZ26, a GW9662 derivate, attenuated obesity-related breast cancer progression by inhibiting the reprogramming of mature adipocytes into to cancer associate adipocyte-like cells

Project description:Obesity has been associated with the development of 13 different types of cancers, including breast cancer. Evidence has indicated that cancer-associated adipocytes (CAAs) promote the proliferation, invasion, and metastasis of cancer. However, the mechanisms that link CAAs to the progression of obesity-related cancer are still unknown. Here, we found the mature adipocytes in the visceral fat of HFD-fed mice have a CAAs phenotype but the stromal vascular fraction (SVF) of the visceral fat has not. Importantly, we found the derivate of the potent PPARg antagonist GW9662, BZ26 inhibited the reprogramming of mature adipocytes in the visceral fat of HFD-fed mice into CAA-like cells and inhibited the proliferation and invasion of obesity-related breast cancer. Further study found that it mediated the browning of visceral, subcutaneous and perirenal fat and attenuated inflammation of adipose tissue and metabolic disorders. For the mechanism, we found that higher PPARg expression was associated with poorer overall survival of breast cancer patients, and further study showed that BZ26 bound and inhibited PPAR by acting as a new modulator. Therefore, BZ26 serves as a novel modulator of PPARg activity that is capable of inhibiting obesity-related breast cancer progression by inhibiting of CAA-like cell formation, suggesting that inhibiting the reprogramming of mature adipocytes into CAAs or CAA-like cells may be a potential therapeutic strategy for obesity-related cancer treatment.

Conditioned media from human pancreatic cancer cells reduces oleic acid oxidation in human myotubes

Project description:In the present study we wanted to metabolically compare healthy primary human pancreas epithelial cells (hPEC) to a pancreatic cancer cell line (PANC-1) and to explore the effect of exposing conditioned medium from the hPEC and PANC-1 cells on the energy metabolism in primary human myotubes. We studied differences in metabolism of glucose, oleic acid and lactic acid, as well as lactic acid production in the two pancreatic cell lines. Mass spectrometry-based proteomics was used to study global protein secretion from the two cell types and pathway analyses were performed. We observed that the PANC-1 cells did not exhibit a typical Warburg effect. Though they tended to have higher uptake of glucose and higher production of lactic acid, they also displayed a higher oxidation of glucose compared to hPEC cells. Interestingly, the PANC-1 cells had higher uptake but lower oxidation of oleic acid, and the mitochondrial reserve capacity from oleic acid was also lower in the PANC-1 cells. These changes in energy metabolism were reflected by differences in gene expressions, and differential expression of different pathways. Furthermore, conditioned media from the PANC-1 cells reduced the oleic acid oxidation in primary human skeletal muscle cells, but further studies will have to be conducted to reveal the mediator of this effect. Although this study is comparing a cancer cell line with a non-cancer cell line, we do not in any way imply that this represent cancer. This study merely represents the differences between the two different cell types studied.
