Project description:Background: Neonicotinoids, a widely used class of insecticide that used is agriculture to control insect’s population, are serious public concern. Objectives: In this study, we examined the capacity of a neonicotinoid, thiacloprid (thia) to induce transgenerational effects in the male mice. Methods: The pregnant outbred Swiss female mice were exposed to thia at embryonic days E6.5 to E15.5 using “0”, and “6” mg/kg/day doses. The genome-wide analysis of DNA methylation analysis showed that differentially methylated regions (DMRs) are located at the promoters and enhancers of many important developmental genes. Notably, DMRs were largely overlapping the germ cell reprograming responsive gene (GRRs) and super-enhancer elements found to be active in ES cells, testis or brain.

Epigenetic Effects Promoted by Neonicotinoid Thiacloprid Exposure.

Project description:BackgroundNeonicotinoids, a widely used class of insecticide, have attracted much attention because of their widespread use that has resulted in the decline of the bee population. Accumulating evidence suggests potential animal and human exposure to neonicotinoids, which is a cause of public concern.ObjectivesIn this study, we examined the effects of a neonicotinoid, thiacloprid (thia), on the male reproductive system.MethodsThe pregnant outbred Swiss female mice were exposed to thia at embryonic days E6.5 to E15.5 using "0," "0.06," "0.6," and "6" mg/kg/day doses. Adult male progeny was analyzed for morphological and cytological defects in the testes using hematoxylin and eosin (H&E) staining. We also used immunofluorescence, Western blotting, RT-qPCR and RNA-seq techniques for the analyses of the effects of thia on testis.ResultsWe found that exposure to thia causes a decrease in spermatozoa at doses "0.6" and "6" and leads to telomere defects at all tested doses. At doses "0.6" and "6," thia exposure leads to an increase in meiotic pachytene cells and a decrease in lumen size, these changes were accompanied by increased testis-to-body weight ratios at high dose. By using RNA-seq approach we found that genes encoding translation, ATP production, ATP-dependent proteins and chromatin-modifying enzymes were deregulated in testes. In addition, we found that exposure to thia results in a decrease in H3K9me3 levels in spermatocytes. The changes in H3K9me3 were associated with a dramatic increase in activity of retroelements.ConclusionOur study suggests that gestational exposure to thia affects epigenetic mechanisms controlling meiosis which could lead to deleterious effects on male spermatogenesis.

Fitness Consequences of Advanced Ancestral Age over Three Generations in Humans.

Project description:A rapid rise in age at parenthood in contemporary societies has increased interest in reports of higher prevalence of de novo mutations and health problems in individuals with older fathers, but the fitness consequences of such age effects over several generations remain untested. Here, we use extensive pedigree data on seven pre-industrial Finnish populations to show how the ages of ancestors for up to three generations are associated with fitness traits. Individuals whose fathers, grandfathers and great-grandfathers fathered their lineage on average under age 30 were ~13% more likely to survive to adulthood than those whose ancestors fathered their lineage at over 40 years. In addition, females had a lower probability of marriage if their male ancestors were older. These findings are consistent with an increase of the number of accumulated de novo mutations with male age, suggesting that deleterious mutations acquired from recent ancestors may be a substantial burden to fitness in humans. However, possible non-mutational explanations for the observed associations are also discussed.

Automated Photochemically Induced Method for the Quantitation of the Neonicotinoid Thiacloprid in Lettuce.

Project description:In this work, we present an automated luminescence sensor for the quantitation of the insecticide thiacloprid, one of the main neonicotinoids, in lettuce samples. A simple and automated manifold was constructed, using multicommutated solenoid valves to handle all solutions. The analyte was online irradiated with UV light to produce a highly fluorescent photoproduct (?exc/?em = 305/370 nm/nm) that was then retained on a solid support placed in the flow cell. In this way, the pre-concentration of the photoproduct was achieved in the detection area, increasing the sensitivity of the analytical method. A method-detection limit of 0.24 mg kg-1 was achieved in real samples, fulfilling the Maximum Residue Limit (MRL) of The European Union for thiacloprid in lettuce (1 mg kg-1). A sample throughput of eight samples per hour was obtained. Recovery experiments were carried out at values close to the MRL, obtaining recovery yields close to 100% and relative standard deviations lower than 5%. Hence, this method would be suitable for routine analyses in quality control, as an alternative to other existing methods.

Residual ground-water levels of the neonicotinoid thiacloprid perturb chemosensing of Caenorhabditis elegans.

Project description:This study investigated the neurological effects of residual ground-water levels of thiacloprid on the non-target organism Caenorhabditis elegans. Nematodes treated with thiacloprid showed a dose-dependent and significantly increased twitch response at concentrations above 50 ng mL-1 that disabled their forward locomotion in liquid culture. In comparison with untreated controls, 10 ng mL-1 thiacloprid perturbed the chemosensory ability of C. elegans such that the nematodes no longer demonstrated positive chemotaxis towards a NaCl chemo-attractant, reducing their chemotaxis index from +0.48 to near to zero. Nematodes also exhibited a locomotion characteristic of those devoid of chemo-attraction, making significantly more pirouetting turns of ≥90° than the untreated controls. Compared to the untreated controls, expression of the endocytosis-associated gene, Rab-10, was also increased in C. elegans that had developed to adulthood in the presence of 10 ng mL-1 thiacloprid, suggesting their active engagement in increased recycling of affected cellular components, such as their nAChRs. Thus, even residual, low levels of this less potent neonicotinoid that may be found in field ground-water had measurable effects on a beneficial soil organism which may have environmental and ecological implications that are currently poorly understood.

Molecular effects of neonicotinoid thiacloprid in honey bees (Apis mellifera)

Project description:The low and high concentrations led to significant alteration of 5 and 71 genes, respectively, of which more were up- than down-regulated. Genes belonging to diverse pathways were affected. KEGG analysis showed that in the high exposure concentration ribosome proteins, mitochondrial oxidative phosphorylation and pyrimidin metabolism were among the three most prominent altered pathways.
