Inducible keratinocyte-specific Mettl3-deficient mice develop spontaneous skin lesions

ABSTRACT: We conducted RNA sequencing analysis on skin tissues obtained from spontaneous inflammatory skin lesions in keratinocyte-specific Mettl3 knockout (K14CreERT2;Mettl3fl/fl) mice and their controls (Mettl3fl/fl). In brief, we observed spontaneous skin inflammation in the tail of K14CreERT2;Mettl3fl/fl mice 2-3 weeks following tamoxifen injection. The objective of this study was to investigate the impact and underlying mechanism of Mettl3 in epidermal keratinocytes regarding skin inflammation

Project description:We analyzed epidermal samples from keratinocyte-specific knockout Mettl3 (K14CreERT2;Mettl3fl/fl) mice and their controls (Mettl3fl/fl) in imiquimod-induced psoriasis-like skin lesions. Briefly, we established an imiquimod-induced psoriasis model in K14CreERT2;Mettl3fl/fl mice and their controls after tamoxifen injection, and epidermal samples were separated and collected from the skin lesions of the mice after 3 or 6 days of modeling (n=3 per group). The aim of this study was to investigate the mechanism by which Mettl3-mediated epidermal m6A methylation affects skin inflammation.

mRNA-sequencing of METTL3-deficient NK cells

Project description:Purpose: For comparing the transcript changes, we conducted mRNA-sequencing of splenic NK cells from Ncr1Cre-Mettl3fl/fl (cKO) and Mettl3fl/fl (WT) mice. Method: Firstly, The splenic NK cells (CD45.2+CD3-NK1.1+NKp46+) are purified via Fluorescence activated Cell Sorting (FACS), then frozen in -80 °C ultra-low temperature refrigerator, followed by High-throughput sequencing, in three replicates, using Illumina Hiseq 1500 platform. Result: Using standard data process workflow, we found the differentailly expressed genes in NK cells from Ncr1Cre-Mettl3fl/fl (cKO) mice, compared with those from Mettl3fl/fl (WT) mice.

Single-cell RNA-seq of embryonic day 17 (E17) mouse skin epithelial cells with or without Mettl3 knockout

Project description:We employed single-cell RNA sequencing to examine effects on steady-state RNA levels induced by METTL3 depletion in mouse skin epithelium during morphogenesis. YFP+ epidermal cells were isolated from E17 embryos with Mettl3 fl/+, Rosa16-YFP fl/+, K14Cre (Ctrl) and Mettl3 fl/fl, Rosa16-YFP fl/+, K14Cre (cKO) genotypes through FACS. Trypan blue staining indicates the ratio of living cells in each sample is >90%. 8000 single cells from each sample is processed with the Chromium™ Single Cell 3’ Library and Gel Bead Kit (V2) (10× Genomics, 120267) to prepare single-cell RNA-seq libraries, which is sequenced on Illumina NextSeq 500 sequenceras 26 × 57 × 8.

Transcriptome-wide maps of m6A RNA methylation marks in murine liver

Project description:N6-methyladenosine (m6A), the most prevalent modification of mRNAs at the (G/A)(m6A)C motif installed by METTL3 that plays critical role in regulation of gene expression. METTL3 is essential for embryonic development, and its dysregulation is linked to various diseases including metabolic disorders and cancer. Here, we provide insight into this by generating Mettl3fl/fl; Alb-Cre (M3LKO) mice carrying a hepatocyte-specific deletion of Mettl3. RNA-seq and MeRIP-seq analyses of mRNA identified significant dysregulation of metabolic genes in M3LKO mice.
