RNA-seq analysis of WT and Tmem11 KO Th1 cells

ABSTRACT: Loss of TMEM11 did not influence T cell differentiation but selectively impaired effector functions of Th1 cells, resulting in reduced severity of symptoms in an animal model of autoimmunity. To find out genes influenced by Tmem11 knockout, we conducted RNA-seq analysis using Th1 cells after re-stimulation.

Project description:We report the differential mRNA expressino of WT and Chi3l1 KO Th1 cells. We cultured WT and Chi3l1 KO naive CD4 T cells under Th1 skewing condition : plate-bound anti-CD3/28 antibody (2ug/mL), IL-12 0.2ng/mL, IL-2 50U/mL, anti-IL-4 neutralizing antibody 2ug/mL, for 3 days. We found different Th1 regulatory and tumoricidal-related gene expression in Chi3l1 KO T cells.

Alcohol intake exacerbates experimental autoimmune prostatitis through activating PI3K/AKT/mTOR pathwaymediated Th1 differentiation

Project description:Epidemiological investigations have revealed a significant association between alcohol consumption and chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS). Nevertheless, the potential mechanisms are still inadequately revealed. This research aimed to investigate the impact of alcohol on CP/CPPS using an animal model and to elucidate the underlying mechanisms. We first established the widely used animal model for CP/CPPS, experimental autoimmune prostatitis (EAP). During the induction of EAP, mice were fed with alcohol or control diet. The HE staining, ELISA, and behavioral experiments were employed to assess the severity of inflammation in EAP mice and EAP-alcohol mice. Patients with a history of chronic alcohol consumption were also included to evaluate the effects of chronic alcohol consumption on CP/CPPS. Subsequently, proteomic analysis, flow cytometry, immunofluorescence, Western blotting, and immunohistochemistry were utilized to investigate the underlying mechanism involved both in vivo and in vitro. HE staining, ELISA, and behavioral experiments showed that alcohol exacerbated the severity of EAP in mice and patients. Proteomic and KEGG pathway analyses showed that abnormal Th1 differentiation and PI3K/AKT/mTOR pathway were significantly enriched. Subsequent mechanistic research showed that alcohol significantly activated PI3K/AKT/mTOR pathway andincreased the Th1 cell differentiation both in vivo and in vitro. In contrast, PI3K inhibitor LY294002 and shRNA-PI3K plasmid inhibited PI3K/AKT/mTOR pathway activation, reduced Th1 cell differentiation, and alleviated EAP inflammation severity, respectively. Our study is the first to demonstrate that alcohol intake promotes Th1 cell differentiation and exacerbates EAP by activating the PI3K/AKT/mTOR pathway. Additionally, the role of LY294002 in inhibiting PI3K/AKT/mTOR pathway to relieve EAP suggests that it can serveas a promising therapeutic target for CP/CPPS.

An intersectional genetic cell depletion unravels the unique role of Th1-Treg cells in tumor immunity

Project description:The immune system comprises various cell types including regulatory T (Treg) cells, and their subsets. Since most of inducible depletion of immune cells for the functional analysis rely on a single protein or gene, it is no longer optimal for the analysis of cell subsets defined by multiple genes. Here we create the Cre- and Flp-double recombinase-dependent intersectional genetic VeDTR mouse system that can selectively label and inducibly deplete a subpopulation of Treg cells called T helper 1 (Th1)-Treg cells. Characterization of Th1-Treg cells using the VeDTR mice revealed the high resistance under oxidative stress, which was required for Th1-Treg cells to accumulate in tumor tissues. Moreover, short-term depletion of Th1-Treg cells only led to anti-tumor immunity but not autoimmunity, whereas that of whole Treg cells did both. Collectively, we have established a novel intersectional genetic immune cell depletion system to reveal unique features of Th1-Treg cells in tumor immunity.

Transcription Factor p73 Regulates Th1 Differentiation

Project description:Inter-individual differences in T helper (Th) cell responses affect susceptibility to infectious, allergic, and autoimmune diseases. To identify factors contributing to these response differences, we analyzed the phenotypic profiles of in vitro-differentiated Th1 cells from 16 inbred mouse strains. Haplotype-based computational genetic analysis indicated that the p53 family protein, p73, affected Th1 differentiation. In cells differentiated under Th1 conditions in vitro, we demonstrated that p73 was a negative regulator of IFNγ production, and this effect required the DNA-binding activity of p73. p73 bound within the gene body or upstream of many genes that affect Th1 differentiation and regulated their expression (e.g., Ifng and Il12rb2). Furthermore, in experimental autoimmune encephalitis, which is a mouse disease model of multiple sclerosis, p73-deficient mice had increased IFNg production and decreased disease severity, whereas in the adoptive transfer model of inflammatory bowel disease, transfer of p73- deficient naïve CD4+ T cells into WT Rag2-/- hosts resulted in increased Th1 responses and enhanced disease severity compared to transfer of WT cells. These studies identify p73 as a regulator of immune responses and indicate that its dysregulation may play a role in susceptibility to autoimmune disease.

Project description:CD4 T cells were activated using CD3/CD28 antibodies in the presence of IL2 and IL12, to generate Th1 cells. Th1 cells were maintained and expanded in IL2 and IL12, on D5 Th1 cells were sorted for CD4+ expression and DAP1 exclusion. After flow sorting, live TH1 cells were resuspended (1e6 per ml) in methionine free RPMI, supplemented with 10% dialysed FBS, IL2 and IL12, and with L-methionine (100μM, or 1μM). Cells were cultured for 5 hrs before collection for proteomics processing.

P2X7 signaling drives Th1 cell differentiation through metabolic reprogramming for aerobic glycolysis

Project description:This study provides evidence on the molecular mechanisms by which P2RX7 signaling promotes Th1 cell differentiation. P2RX7 induces T-bet expression and aerobic glycolysis in splenic CD4+ T cells that respond to malaria, at a time prior to Th1/Tfh polarization. Cell-intrinsic P2RX7 signaling sustains the glycolytic pathway and causes bioenergetic mitochondrial stress in activated CD4+ T cells. We also show in vitro the phenotypic similarities of Th1-polarized CD4+ T cells that do not express P2RX7 and those in which the glycolytic pathway is pharmacologically inhibited. In addition, ATP synthase blockade in vitro and the consequent inhibition of oxidative phosphorylation, which forces cells to use aerobic glycolysis, is sufficient to promote rapid CD4+ T cell proliferation and polarization to the Th1 profile in the absence of P2RX7. These data demonstrate that P2RX7-mediated metabolic reprograming for aerobic glycolysis is a key event for Th1 cell differentiation and suggest that ATP synthase inhibition is a fundamental mechanism by which P2X7 signaling induces the Th1 response.

We will also define the role of IL-17 signaling in the pathology by comparing cells derived from wild type mice to those derived from mice lacking Act1, a key adaptor required for IL-17 dependent signaling. Brains and spinal cords from animals at peak of disease will be profiled.
