Gene expression in Drosophila treated with P. nigrum, pyrethrin or P. nigrum plus pyrethrin

ABSTRACT: The purpose of this experiment was to assess the effect of a synergistic combination of natural pyrethrin and an ethylacetate extract of Piper nigrum seeds (a botanical insecticide). This effect was compared to the effects of P. nigrum or pyrethrin used alone. Due to the synergistic nature of the mixture, it was predicted that gene expression profiles in this treatment would reflect this effect. Keywords: insecticide response, stress-response

Project description:This experiment was used to determine the effect of a botanical insecticide upon gene expression profiles in Drosophila melanogaster. Adult female Drosophila (oregon-R strain) were treated with an ethylacetate extract of Piper nigrum (Piperaceae) seeds formulated in 99% ethanol. Treatment was topical, using a Potter's tower to administer a total of 2 mL of a 0.9mg/mL concentration. Control treatment was identical except flies were treated with 99% ethanol as a solvent control. Gene expression was studied four hours post-treatment. Effect of water stress combined with sulfur deficiency on reproductive processes and seed composition in pea

Project description:). In this study we describe the effect of drought combined or not with S-deficiency on pea productivity, nutrient partitioning between plant parts and seed quality traits. A moderate water stress was applied at the beginning of the reproductive phase, a period during which drought frequently occurs in the field. Sharp differences in the plant and seed characteristic in response to the combined stress compared to individual stresses were observed, highlighting synergistic effects on reproductive processes and antagonistic effects on seed protein composition, which could be explained by changes in the sink strength of the seeds for nitrogen.

Grapevine response to a Dittrichia viscosa extract and a Bacillus velezensis strain

Project description:The present study aims to evaluate the response of the three Mediterranean local grapevines ‘Garnacha Blanca’, ‘Garnacha Tinta’, and ‘Macabeo’ to treatments with biocontrol products (BPs), a botanical extract (Akivi, Dittrichia viscosa extract) and a beneficial microorganism (Bacillus UdG, Bacillus velezensis). A combination of transcriptomics and metabolomics approaches were chosen in order to study grapevine gene expression and to identify gene marker candidates, as well as, to determine grapevine metabolites differentially concentrated in response to BPs treatments. Grapevine plants were cultivated in greenhouse controlled conditions and submitted to the treatments, and thereafter, leaves were sampled 24h after treatment to conduct gene expression study by RNA-sequencing for ‘Garnacha Blanca’ leaves extract and by RT-qPCR for the three cultivars. Differentially expressed genes (DEGs) were investigated for both treatments and highly influenced DEGs were selected to be tested in the three cultivars as treatment gene markers. In addition, extraction of leaf components was performed to quantify metabolites such as phytohormones, organic acids, and phenols. Considering all the upregulated and downregulated genes and enhanced metabolites concentrations, the treatments had an effect on jasmonic acid, ethylene, and phenylpropanoids defense pathways. In addition, several DEG markers were identified presenting a stable overexpression after the treatments in the three grapevine cultivars. These gene markers could be used to monitor the activity of the products in field treatments in future research. Further research will be necessary to confirm these first results under field conditions.

Molecular effects of 1-naphthyl-methylcarbamate and solar radiation exposures on human melanocytes

Project description:Carbaryl (1-naphthyl-methylcarbamate), a broad-spectrum insecticide, has recently been associated with the development of cutaneous melanoma in an epidemiological cohort study with U.S. farm workers also exposed to ultraviolet radiation, which is known to be the main etiologic factor for skin carcinogenesis. Although comprehensive and well designed, the agricultural epidemiological study was not sufficient to characterize the direct contribution of the insecticide and solar radiation in melanomagenesis. Several studies have explored the synergistic effect of certain chemicals with UV radiation, increasing its deleterious effects on the skin. We hypothesized that carbaryl exposure associated with UV solar radiation may induce melanocyte transformation. This study aimed to characterize human melanocytes after individual or combined exposure to carbaryl (100 μM) and solar radiation (375 mJ/cm2). In a microarray analysis, carbaryl, but not solar radiation, induced an important oxidative stress response, evidenced by the upregulation of antioxidant genes, such as Hemeoxygenase-1 (HMOX1), and downregulation of Microphtalmia-associated Transcription Factor (MITF), the main regulator of melanocytic activity; results were confirmed by qRT-PCR. Carbaryl and solar radiation induced a gene response suggestive of DNA damage and cell cycle alteration. The expression of genes in these categories was notably more intense in the combined treatment group, in a synergistic manner, for CDKN1A, BRCA1/2 and MDM2 genes. Likewise, flow cytometry assays demonstrated S-phase cell cycle arrest, reduced apoptosis levels and faster induction of cyclobutane pyrimidine dimers (CPD) lesions in carbaryl treated groups. Our data suggests that carbaryl is genotoxic to human melanocytes, especially when associated with solar radiation.
