ABSTRACT: we evaluated the modulation of gene expression profile in PBMCs from patients with hypotalamic amenorrhea. We have employed whole genome microarray expression profiling as a discovery platform to identify genes differentially expressed upon metreleptin treatment at different time points, week 12, 24 and 36.

Project description:PBMCs from major depressive episode (MDE) subjects were obtained at two time points: during a severe episode of depression and eight week later after clinical remission. PBMCs from sex- and age-machted controls were also obtained at an eight weeks interval. PBMC total RNAs from MDE and control subjects were profiled after hybridization with Agilent SurePrint G3 Human GE 8x60K Microarray to identify blood biomarkers of MDE. Total RNAs were extracted from 36 samples including: 9 control and 9 MDE subjects at two point of time (first and eight week later)

Human-derived PBMCs were stimulated by different groups to kill tumor cells.After 48 hours, PBMCs were taken out, and the enrichment of activated proteins of relevant lymphocytes and the regulation of pathways were examined by proteomics.

Project description:In this study, proteomics was used to explore the activation of lymphocytes and related pathways in PBMCs of different stimulation groups. In our study, proteomics analysis showed that the immune cells in PBMCs activated by stimulation group exhibit improved functions related to white blood cell activation, proliferation and cytotoxicity during biological processes. At the same time, KEGG pathway analysis showed that the signaling pathways’ key proteins for leukocyte transendothelial migration, Th1/Th2 cell differentiation, JAK-STAT (involved in many important biological processes such as cell proliferation, differentiation, apoptosis and immune regulation), DNA replication and Th17 cell differentiation were up-regulated; whereas the key proteins of necroptosis pathways in immune cells were down-regulated.

Project description:The project aims to measure targeted and non-targeted metabolite data of intracellular extracts of uninfected and Leishmania-donovani infected macrophages at 0, 12, 36 and 72 hours post infection using a multiplatform mass spectrometry approach combining CE-TOF/MS (polar metabolites), LC-QTOF/MS (non-polar metabolites) and LC-QqQ/MS (polar metabolites) to characterize the dynamics of metabolic alterations ocurring in the human macrophage upon L. donovani infection.
