Project description:Genome-Scale draft model for Human Peripheral Blood Mononuclear Cells (PBMCs). A GEM for PBMCs was developed by applying the INIT algorithm on Human Metabolic Reconstruction (HMR 2.0) as a template model. GEMs were contextualised/ constrained for different conditions using expression datasets. The gene/transcript expression data obtained from PBMCs of Type 1 Diabetes progressors, non-progressors, and healthy controls were employed to score each reaction of HMR 2.0. Rhesus macaque model of hCoV-EMC respiratory infection

Project description:Microarray analysis of peripheral blood mononuclear cells (PBMCs), lungs, and lung lesions collected over the course of hCoV-EMC infection of 6 rhesus macaques. 6 rhesus macaques were infected intratracheally with hCoV-EMC. PBMCs were collected at days 0, 1, 3, and 6, and lungs were collected from serial sacrifices of 3 animals each at day 3 and day 6. Infection produced a mild-moderate, self-limiting respiratory infection, and was not lethal. We performed microarray analysis (using Agilent Rhesus arrays) on all lungs, lung lesions, and PBMCs collected for the study.

Project description:Elderly atopic dermatitis is a subtype of AD defined by age (over 60 years), which has different clinical manifestations and distinct inflammatory patterns from AD in infants, children, and adolescents/adults. However, the molecular characteristics of elderly AD remain to be clarified.We sought to characterize the molecular features of skin lesions and peripheral blood mononuclear cells (PBMCs) in patients with AD across various age groups, focusing on elderly AD.We identified the molecular phenotypes of skin lesions and PBMCs in elderly individuals with AD. Fibroblasts, innate immune cells, and SASP might play important roles in the pathogenesis of elderly AD.

Gene Expression Profiling in Peripheral Blood in Untreated New Onset Systemic Juvenile Idiopathic Arthritis

Project description:Systemic Juvenile Idiopathic Arthritis (sJIA) has been strongly associated with macrophage activation syndrome (MAS). To better understand the pathogenesid of sJIA and to facilitate the search for MAS biomarkers, we examine gene expression profiles in untreated new onset sJIA. 17 new onset sJIA patients were included in the study. 5 of the 17 patients showed evidence of subclinical MAS and 2 eventually developed overt MAS. Keywords: disease versus control Peripheral blood mononuclear cells (PBMCs) were separated using a Ficoll gradient from the 17 new onset sJIA patients and 30 normal control. RNA was extracted from the PBMCS and subsequently hybridized to Affymetrix microarrays
