Project description:Transcriptional regulator REST plays a key role in repressing neuronal specific genes in prostate cancer and other non-neuronal tissues. Moreover, loss of REST is observed in neuroendocrine prostate tumors. Here, we use ChIP-seq analysis to study genome–wide REST occupied regions in the prostate cancer cell line, LNCaP. REST occupied regions were then correlated to gene expression changes occurring between prostate adenocarcinoma and neuroendocrine prostate tumors in vivo.

siRNA profiling of human LNCaP cells overexpressing/knockdown of HOXC6

Project description:The transcription factor HOXC6 is upregulated in human prostate cancer. SiRNA knockdown of HOXC6 induces apoptosis in LNCaP cells while upregulation rescued LNCaP cells from siRNA-induced apoptosis. We used microarrays to identify the genes whose expression underly the anti-apoptotic and proliferative effects of HOXC6 in LNCaP cells. Experiment Overall Design: LNCaP cells were transiently transfected with a cocktail of 5 siRNAs to HOXC6 to a final concentration of 100nM, with a HOXC6 gene expression vector, or control siRNA and vector. We used microarrays to detail the global changes of gene expression when LNCaP cells were treated with siRNAs against SLC45A3-ELK4. We transfected LNCaP cells with siRNAs against SLC45A3-ELK4 and siGl2 control. 72hrs later, RNAs were harvested.
