Chronic Salmonella infected mouse model.

Project description:The bacterial infected mouse model is a powerful model system for studying areas such as infection, inflammation, immunology, signal transduction, and tumorigenesis. Many researchers have taken advantage of the colitis induced by Salmonella typhimurium for the studies on the early phase of inflammation and infection. However, only few reports are on the chronic infection in vivo. Mice with Salmonella persistent existence in the gastrointestinal tract allow us to explore the long-term host-bacterial interaction, signal transduction, and tumorigenesis. We have established a chronic bacterial infected mouse model with Salmonella typhimurium colonization in the mouse intestine over 6 months. To use this system, it is necessary for the researcher to learn how to prepare the bacterial culture and gavage the animals. We detail a methodology for prepare bacterial culture and gavage mice. We also show how to detect the Salmonella persistence in the gastrointestinal tract. Overall, this protocol will aid researchers using the bacterial infected mouse model to address fundamentally important biological and microbiological questions.

Plasma metabolomics of diverse mouse strains infected with Plasmodium chabaudi

Project description:To uncover links between metabolism and disease severity in murine malaria, we performed plasma metabolomics via Metabolon on eight inbred, Plasmodium chabaudi-infected mouse strains with diverse disease phenotypes. We sacrificed and collected plasma from >=3 mice per strain per day of acute infection alongside uninfected control mice (approximately days 5-12 depending on mouse strain). We collected disease severity data, e.g. weight loss, liver enzymes, and anemia, concurrently. Together, these data enable 1) a picture of strain-specific and conserved metabolic responses during acute malaria, and 2) a comparison between metabolic responses and disease severity.

Dual RNA-seq – sRNA profiling in various cell types

Project description:Bacteria possess many small noncoding RNAs whose regulatory roles in pathogenesis are little understood due to a paucity of macroscopic phenotypes in standard virulence assays. Here, we use a novel Dual RNA-seq approach for a single-step simultaneous RNA profiling in both pathogen and host to reveal molecular phenotypes of sRNAs during infection with Salmonella Typhimurium. We identify a new PhoP/Q-activated small RNA which upon bacterial internalization acts to temporally control the expression of both, invasion-associated effectors and virulence genes required for intracellular survival. This riboregulatory activity is shown to adjust the human response to replicating Salmonella, and have a pervasive impact on host RNA expression both inside and outside protein-coding regions including infection-specific alterations of an array of long noncoding RNAs. Our study provides a paradigm for a comprehensive RNA-based analysis of intracellular bacterial pathogens without their physical purification from a host and a new discovery route for hidden functions of pathogen genes.

Mouse melanoma types with various genotypes

Project description:Only a subset of melanoma patients has evidence for spontaneous T cell infiltration into tumor sites, previously associated with clinical responses. However, the molecular mechanisms explaining absence of a T cell response are not yet defined. Analyses of human melanoma metastases have revealed that T cell signature low tumors show alterations in the Wnt/b-catenin signaling pathway. We utilized an inducible mouse model driven by inducible BrafV600E and PTEN-deletion, with or without active b-catenin (CAT-STA) to test if tumor intrinsic active b-catenin can block anti-tumor immunity. While Braf/PTEN melanomas showed presence of a T cell infiltrate, T cells were nearly completely eliminated in tumors expressing active b-catenin. Adoptive transfer experiments revealed defective T cell priming when tumors expressed active b-catenin. Analysis of the antigen-presenting cell compartment revealed a selective decrease in the CD103+ DC subset within the tumor microenvironment, which could be associated with b-catenin depended inhibition of expression of the chemokine CCL4 within tumor cells. Here we used 3 mice of each genotype BrafV600E/PTEN-/- and BrafV600E/PTEN-/-/Cat-STA and compared their gene-expression profiles.
