Response to LiCl of galactose grown cells

Project description:Wild type strain CEN.PK113-7D was grown in an aerobic batch cultivation with a start concentration of 20 g/L galactose. During exponential growth at a biomass concentration of 3 g dry weight/L LiCl was added to a concentration of 10 mM. Just before addition of LiCl (time 0) and 20, 40, 60 and 140 minutes after addition of LiCl samples were taken for transcription analysis. Lithium inhibits phosphoglucomutase whereby both galactose uptake and growth is strongly affected. Keywords = Saccharomyces galactose lithium Keywords: time-course

Regulation of gene expression by Hcm1 in chronic LiCl stress

Project description:Gene expression was quantified in S. cerevisiae cells after 40 hours of LiCl treatment or control conditions. Cells expressing wild type (WT) Hcm1, a stable Hcm1 mutant (Hcm1-3N), or a consitutitvely active, phosphomimetic mutant (Hcm1-8E) were compared to allow the identification of genes whose expression depends upon dynamic phosphorylation of Hcm1. We found that dynamic phosphorylation of Hcm1 is required for expression of Hcm1 target genes in LiCl stress.

N2 worms treated with Resveratrol

Project description:Eggs were isolated by hypochlorite treatment and allowed to hatch in the absence of food as L1 stage arrested worms in S-buffer. Worms were then transferred to large plates containing NGM alone or NGM+ resveratrol and allowed to grow till young adult stage (ca 40 hours). Worms were harvested and total RNA extracted using Trizol reagent. Poly-A+ RNA was isolated using Qiagen midi-prep kit and used for microarray hybridization.
