A Surgical Method for Continuous Intra-Portal Infusion of Gut Microbial Metabolites in Mice

Project description:Gut-derived microbial metabolites have been shown to play key roles in human physiology and disease. However, establishing mechanistic links between gut microbial metabolites and disease pathogenesis in animal models presents many challenges. The major route of absorption for microbe-derived small molecules is venous drainage via the portal vein to the liver. In the event of extensive liver first pass- or presystemic hepatic metabolism, the route of administration of these metabolites becomes critical. Here we describe a novel portal vein cannulation technique using a subcutaneously implanted osmotic pump to achieve continuous portal vein infusion in mice. First, the microbial metabolite trimethylamine (TMA) was administered over 4 weeks and compared to a vehicle control. Using a liquid chromatography-tandem mass spectrometry (LC-MS/MS), an increase in peripheral plasma levels of TMA and its host liver-derived co-metabolite trimethylamine-N-oxide (TMAO) were observed in a sexually-dimorphic manner. Next, 4-hydroxyphenylacetic acid (4-HPAA), a structurally-distinct microbial metabolite that undergoes extensive hepatic first pass metabolism, was administered portally to examine its effects on hepatic gene expression. Peripheral plasma levels showed no difference in free or total 4-HPAA. However, while liver tissue demonstrated no difference in free 4-HPAA, total levels were increased when compared to the control group. More importantly, significant changes were observed in hepatic gene expression using an unbiased RNA sequencing approach. Collectively, this work describes a novel method for administering gut microbe-derived metabolites via the portal vein, mimicking their physiologic delivery in vivo.

Identification of a portal vessel-associated fetal liver hematopoietic stem cell niche

Project description:Whereas the cellular basis of the hematopoietic stem cell (HSC) niche in the bone marrow has been characterized, the nature of the fetal liver (FL) niche is not yet elucidated. We show that Nestin+NG2+ pericytes associate with portal vessels, forming a niche promoting HSC expansion. Nestin+NG2+ cells and HSCs scale during development with the fractal branching patterns of portal vessels, tributaries of the umbilical vein. After closure of the umbilical inlet at birth, portal vessels undergo a transition from Neuropilin-1+Ephrin-B2+ artery to EphB4+ vein phenotype, associated with a loss of peri-portal Nestin+NG2+ cells and emigration of HSCs away from portal vessels. These data support a model in which HSCs are titrated against a peri-portal vascular niche with a fractal-like organization enabled by placental circulation.

Tryptophan metabolites in mouse serum

Project description:Analyzing murine sera to investigate into which tryptophan metabolites in the host circulation are derived from diet, host metabolism and/or the gut microbiome. Blood was collected from the portal vein of conventional C567BL/6J mice and sera analyzed by targeted LC-MS. To test this hypothesis and evaluate the necessity of microorganisms to produce these examined tryptophan metabolites, sera were collected from germ-free (GF) mice and subjected to the same targeted metabolomic analysis. Having generated Trp metabolite profiles from sera, we asked if altering the diet would significantly change circulating concentrations. One of two groups of conventional C57BL/6J mice with ad libitum access to a standard laboratory chow were switched onto a semi-purified diet (AIN-93G) while the second remained on standard chow. After one week, portal vein blood was collected and analyzed using targeted LC-MS.

The COPPER Trial: Comparision of a tumour marker in patients with appendix or right sided colon cancer

Project description:Interventions: The only intervention will be to take five ml of blood will be taken from a vein in the patient’s arm (this will be from a intravenous cannula as part of the patient’s general anaesthetic such that there will be no additional venepuncture burden) as well as another 5 ml of blood from the ileocolic / appendicular vein (a blood vessel that drains blood from the appendix or right colon that is located in the right lower side of the abdomen) during the initial stages of your peritonectomy surgery (total number of blood tests: 2; total volume: 10ml; NB: 5-10mL is equivalent to 1-2 teaspoons). The blood sampling would take less than one minute during surgery by the anaesthetist (peripheral sample) and the surgeon (portal vein sample). Assessment of the fidelity of this intervention will be in part by testing for albumin of these specimens for comparison. Project description:We report a tissue-derived KLRG1-ILC2s highly expressing CD69 in HCC, which are distinguished from circulating ones from portal vein and peripheral blood. Relief of KLRG1 inhibitory signaling, the HCC-induced ILC2s tend to shape an immunosuppressive tumor microenvironment through CXCL2/neutrophil axis. Furthermore, the abundance of ILC2s in HCC correlated with the recurrence and progression free survival (PFS) of HCC patients indicated ILC2s being an emerging target for HCC immunotherapy.

Transcription profiling of superior mesenteric arteries from rats that have had portal vein ligation or a sham procedure for different lengths of time

Project description:Twenty-four male Sprague-Dawley rats were used for this experiment, a partial portal vein ligadure (PVL) was carried out in 12 rats, the other 12 rats were sham-operated. Superior mesenteric artery (SMA) samples were sequentially obtained from 2 rats from the PVL and sham groups at 1, 6, and 24 hours, and 3, 5 and 14 days after portal vein ligation or sham procedure. Each PVL sample was paired with its corresponding sham sample using a dye swap experiment design (technical replicates) resulting in 12 arrays (6 time points x 2 dye swap).
