Glomerular tissue extra-cellular proteome in focal segemental glomerular sclerosis

ABSTRACT: Glomeruli were isolated from formalin-fixed paraffin embedded (FFPE) preserved human kidney biopsies using laser capture micro-dissection. Isolated tissue was de-cellularized by extraction using an ammonium hydroxide/NP-40 protocol. The residual extra-cellular fraction was analyzed by 1-dimensional C18 RP LCMS.

Comparison of Glomerular and Podocyte mRNA Profiles in eNOS-/- Diabetic Mice Induced by Streptozotocin

Project description:We compared mRNA profiles of isolated glomeruli versus sorted podocytes between diabetic and control mice. IRG mice crossed with eNOS-/- mice were further bred with podocin-rTTA and TetON-Cre mice to permanently label podocytes before the diabetic injury. Diabetes was induced by injection of streptozotocin. mRNA profiles of isolated glomeruli and sorted podocytes from diabetic and control mice at 10 weeks after induction of diabetes were examined. Consistent with the previous reports, expression of podocyte-specific markers in the glomeruli were down-regulated in the diabetic mice compared to controls. However, these differences disappeared when mRNA levels were corrected for podocyte number per glomerulus. Interestingly, the expression of these markers was not altered in sorted podocytes from diabetic mice, suggesting that the reduced expression of podocyte markers in isolated glomeruli is likely a secondary effect of reduced podocyte number, rather than the loss of differentiation markers. Analysis of the differentially expressed genes in diabetic mice also revealed distinct up-regulated pathways in the glomeruli (mitochondrial function and oxidative stress) and podocytes (actin organization). In conclusion, our data suggest that podocyte-specific gene expression in transcriptome obtained from the whole glomeruli may not represent those of podocytes in the diabetic kidney. Podocyte-specific deletion of tubular sclerosis complex 2 causes focal segmental glomerular sclerosis and progressive renal failure

Project description:Obesity can initiate and accelerate the progression of kidney diseases. However, it remains unclear how obesity affects renal dysfunction. Here, we show that a newly generated podocyte-specific tubular sclerosis complex 2 (Tsc2) knockout mouse model (Tsc2∆podocyte) develops proteinuria and dies due to end-stage renal dysfunction by 10 weeks of age. Tsc2∆podocyte mice exhibit an increased glomerular size and focal segmental glomerulosclerosis, including podocyte foot process effacement, mesangial sclerosis and proteinaceous casts. Podocytes isolated from Tsc2∆podocyte mice show nuclear factor, erythroid derived 2, like 2-mediated increased oxidative stress response on microarray analysis and their autophagic activity is lowered through the mammalian target of rapamycin (mTOR)—unc-51-like kinase 1 pathway. Rapamycin attenuated podocyte dysfunction and extends survival in Tsc2∆podocyte mice. Additionally, mTOR complex 1 (mTORC1) activity is increased in podocytes of renal biopsy specimens obtained from obese patients with chronic kidney disease. Our work shows that mTORC1 hyperactivation in podocytes leads to severe renal dysfunction and that inhibition of mTORC1 activity in podocytes could be a key therapeutic target for obesity-related kidney diseases.

Project description:＜Project description＞ Data independent acquisition (DIA) mass spectrometry of Isolated glomeruli from homozygous deletion of the nuclear export signal of p62 (dNES) and WT mice (n=3 each). ＜Sample processing＞ Formalin fixed paraffin embedded (FFPE) tissues were prepared in general method and kidney samples were cut into 5µm-thick sections on the membrane slides (Leica Microsystems, Mannheim, Germany). Glomeruli were isolated via laser capture micro-dissection (0.6 mm2 in total). The dissected glomeruli were collected separately into 40 µL of Tris EDTA buffer and 0.002% Zwittergent 3-16 detergent.　Solubilised proteins were further digested into peptides by 0.01ug/ul trypsin (Promega, Madison, WI, USA) overnight. Each sample was centrifuged at 15000 rpm for 30 minutes prior to measurement. ＜Mass spectrometry and data processing＞ Samples were measured by DIA-MSMS using NanoLC-MSMS system with Vanquish Neo and Orbitrap Exploris 240 (Thermo Scientific). The measurement data were analyzed with DIA-NN 1.8.1.

Gene expression analysis of rat glomerular parietal epithelial cells

Project description:Very little is known about the function of glomerular parietal epithelial cells (PECs). In this study, we performed genome-wide expression analysis on PEC-enriched capsulated vs. PEC-deprived decapsulated rat glomeruli to determine the transcriptional state of PECs under normal conditions. We identified hundreds of differentially expressed genes that mapped to distinct biologic modules including development, tight junction, ion transport, and metabolic processes. Since developmental programs were highly enriched in PECs, we characterized several of their candidate members at the protein level. Collectively, our findings confirm that PECs are multifaceted cells and help define their diverse functional repertoire. We developed a sequential sieving methodology to selectively isolate capsulated (PEC-enriched) glomeruli from un-capsulated (PEC-deprived) glomeruli in rats. Three samples were obtained for each preparation. Total RNA was isolated and its integrity confirmed via Bioanalyzer 2100. Each sample (n =6) was hybridized to an Affymetrix Rat GeneChip 1.0ST microarray.
