Proteome and phosphoproteome of brassinolide treated Arabidopsis

ABSTRACT: We performed proteome and phosphoproteome profling of Arabidopsis plants to determine brassinosteroid responsive changes. Seeds were germinated and grown for 7 days on plates containing 1 uM brassinozole (BRZ) to eliminate any background BR signaling. on day 7 plants were treated with brassinolide (0 uM or 1 uM BL) for six different lengths of time (15min, 30min, 1hr, 2hr, 4hr, 8hr). Proteins were extracted and digested using the Phenol-FASP method described in https://doi.org/10.1002/pmic.201800220. Peptides were TMT labeled. Phosphopeptides were serially enriched using High-Select TiO2 and High-Select Fe-NTA kits.
