FLAG-BIo-tagged MAT2A interactome analysis in HeLa cells

ABSTRACT: MAT2A is the enzyme synthesising S-adenosylmethionine which is utilized as the primary methyl group donor in biological methylation.HeLa cells that stably expressed the FLAG-Bio-tagged mouse MAT2A protein were established by retroviral infection. The FLAG-Bio-MAT2A cells, as well as normal HeLa cells as control, were lysed and the lysates were subjected to affinity purification using streptavidin to isolate FLAG-Bio-MAT2A together with its interacting proteins. The isolated proteins were separated by SDS-PAGE, and every lane was divided into 3 gel pieces. Then the proteins in the gel pieces were alkylated using acrylamide, were in-gel trypsin-digested and were subjected to LC-MS/MS for protein identification. Every 3 raw data files derived from the same sample were converted into a single MGF file before the database search. Raw files 200110-1 to 20110-3: control cell purification, files 200110-4 to 20110-6: FLAG-Bio-MAT2A cell purification.
