Immunoprecipitation was performed with HCT116 cells overexpressing 3xFLAG-SET/TAF-Ib with anti-FLAG antibodies

ABSTRACT: Immunoprecipitation was performed with HCT116 cells overexpressing 3xFLAG-SET/TAF-Ib with anti-FLAG antibodies. The immunoprecitates were separated by SDS-PAGE and putative interaction partners of SET/TAF-Ib were identified with mass spectrometry.

Two different sources of RNA were analyzed: 1.) HCT116 colon cancer cells overexpressing human mir-30a-5p together with a sponge vector expressing binding sites for the antisense sequence of sh30a-5p vector construct to compete out possible miRNAs derived from the anti-guide strand of this vector. 2.) HCT116 colon cancer cells expressing control vector (pLKO.1-LV) and the sponge vector described above.

Project description:LexA is a transcriptional repressor for genes requiring expression primarily during SOS response in response to stress such as that caused by DNA alkylating agent Mitomycin C (MMC). LexA undergoes self-cleavage under stress conditions thereby lifting the repression on genes whose expression is required for stress response. In our experiment LexA binding sites in the genome were determined in untreated and MMC treated (overnight, 14h) cultures using a LexA-3xFLAG strain and anti-FLAG monoclonal antibodies. Wild type Streptomyces venezuelae (unFLAGged LexA) cultures, with and without MMC treatment were used as negative controls.
