Proteomic and phosphoproteomic characterization of cardiovascular dysfunction arising from exposure to space radiation

ABSTRACT: Mice were exposed to 150 cGy Galactic Cosmic Ray (GCR) versus sham, and hearts and plasma were harvested at 8 months post radiation. Hearts were homogenized in 5% SDS and 250 ugs protein was digested with trypsin using an S-trap followed by TMT labeling, high-pH reversed-phase fractionation and phosphopeptide enrichement. 12 fractions (unenriched and phospho-enriched) were analyzed by nanoLC-MS/MS using an MS2 method. 15 uL plasma was processed using sodium-deoxycholate assisted trypsin digestion followed by microflow LC-MS/MS using data-independent acquisition.
