Gene expression profiling of the ileum following vertical sleeve gastrectomy compared to pair-fed sham surgery controls in mice with high-fat diet-induced obesity.

Project description:We report the expression profiles of ileal samples extracted from obese male C57Bl6 mice following vertical sleeve gastrectomy or sham surgery, focusing of gene signatures indicative of altered bile acid metabolism. We induced obesity in male C57bl6 mice through a high-fat diet. At 8 weeks, they were submitted to either a vertical sleeve gastrectomy (VSG) or a sham surgery; following surgery VSG mice were fed ad libitum while sham controls were pair-fed (SPF) to the experimental animals. During the four perioperative days, mice were fed a liquid Osmolite diet. Approximately 8 weeks after surgery mice were fasted overnight and gavaged with .5mL Osmolite; after one hour mice were sacrificed and the terminal ileum were extracted. RNA was extracted, quantified, and compared between VSG and SPF experimental groups.

Transcriptome profiling of wildtype, nonoperated mouse hearts compared to wildtype, sham operated hearts

Project description:The purpose of the present study was to investigate the activation of the cardiac inflammatory response in the hearts of SHAM (surgical opening of the chest) mice in comparison to CONTROL hearts isolated from naïve (not operated) mice. Male C57BL/6J (10 weeks) were subjected to an open-chest ischemia-reperfusion (I/R) SHAM procedure in order to examine the effects of this surgery alone in producing cardiac inflammation. The procedure consisted of thoracotomy and a suture passed around the left anterior descending artery (LAD). The mouse chest was kept open for approximately 45 min. We chose to study an open-chest I/R SHAM because this is the reference surgery for the most common surgical procedure used to study cardiac inflammation. Mice were sacrificed 5 days after the procedure and the hearts were removed and used in RNAseq experiments.

Partial urethra obstruction in rat

Project description:Urethra was partially ligated and the urinary bladder was removed 10 days or 6 weeks after obstruction. Sham operated rats were used as controls. An addtitonal group of rats were repoerated 6 weeks after surgery and the obstruction was removed. These rats were then sacrificed 10 days after deobstruction. Project description:Purpose: RNAseq analyses were conducted to screen for the genes undergoing transcriptional changes either in the liver of high-fat-diet (HFD)-induced obese mice or in the liver of Lepr-deficient db/db mice compared to the livers of the respective control mice Methods: C57BL/6 wild-type male mice were fed on high-fat diet (HFD) or a low-fat diet (NCD) for 18 weeks starting from 6 weeks of age, and the livers were collected at 24 weeks of age at ad libitum-fed condition.Misty/misty or db/db were sacrificed at ad libitum-fed condition at 10weeks and the liver was collected. Results: 2079 genes and 1085 genes were identified in high-fat-diet fed mice and db/db mice, respectively.

Effects of high-fat induced obesity on gene expression in mouse kidney

Project description:Global gene expression in the 20-week remnant kidneys of uninephrectomized mice fed either a chow or a high fat diet was compared with kidney tissue from the corresponding sham groups. Results provide insight into mechanisms underlying effects of high-fat induced obesity on gene expression in mouse kidney. Male C57/BJ mice aged 6 weeks were randomly assigned to uninephrectomy (UNX) or sham procedures and fed with a high fat diet or control chow diet. All mice were sacrificed under anesthesia 20 weeks after surgery and kidneys were harvested. 3-4 total RNA samples per group were analyzed and gene expression was compared between groups.
