Standardized gnotobiotic mouse model for NMR based phenotyping

ABSTRACT: Germ-free mice were inoculated with 15 representative bacterial strains selected from specific pathogen-free mice. All three microbiota mouse models were generated in two different facility sites and collected plasma sample phenotyped by NMR. 58 mice from Facility 1 (32 males and 26 females) and 59 mice from Facility 2 (29 males and 30 females) were used for analysis.

Gender bias in autoimmunity is influenced by microbiota

Project description:Gender bias and the role of sex hormones in autoimmune diseases are well established. In specific-pathogen free (SPF) non-obese diabetic (NOD) mice females have 1.3-4.4 times higher incidence of Type 1 diabetes (T1D). Germ-free (GF) mice lose the gender bias (female/male ratio 1.1-1.2). Gut microbiota differed in males and females, a trend reversed by male castration, confirming that androgens influence gut microbiota. Colonization of GF NOD mice with defined microbiota revealed that some but not all lineages overrepresented in male mice supported a gender bias in T1D, and protection did not correlate with androgen levels. However, hormone-supported selective microbial lineage variation may work as a positive feedback mechanism contributing to the sexual dimorphism of autoimmune diseases. Gene expression analysis suggested pathways involved in protection of males from T1D by microbiota.

Sex and social status influence DNA methylation dynamics in a polygynous bat

Project description:DNA methylation (DNAm) influences when and where genes are expressed or repressed during growth and development. Recently, patterns of DNAm at conserved genomic sites have been discovered that predict chronological age in all mammals. Deviations from these ‘epigenetic clocks’ have been used to identify factors that alter the aging process. In this study, we profile DNAm with a custom microarray for over 330 wild Phyllostomus hastatus, a highly polygynous bat in which unrelated females form long-lasting associations and larger males compete aggressively to control mating access to female groups. DNAm age estimates reveal that females can live more than 1.5 times as long as males. After fitting linear models for age, sex and their interaction we find that DNAm changes 1.4 times faster in males than females at thousands of sites. Therefore, even though age of either sex can be predicted by a common set of sites, the methylome of males is more dynamic than that of females. Sites associated with differences in the rate of change between males and females are sensitive to androgens and enriched on the X chromosome. Those that gain methylation with age in both sexes are associated with active and repressive chromatin states in blood and are enriched in promoters of genes involved in regulation of metabolic processes. In contrast, few sites differ in DNAm rate between males of different reproductive status, though subordinate males exhibit faster DNAm change than dominant males. Thus, males have accelerated physiological processes in comparison to females that likely increase mating success but reduce survival.

Vitamin C modulates the levels of several proteins of the mitochondrial complex III and its activity in the mouse liver

Project description:In the present study, we used the Gulo-/- female and male mice model which depends entirely on ascorbate derived from the diet. Importantly, the levels of ascorbate found in the serum of Gulo-/- mice reflect the amounts of ascorbate provided in drinking water. We tested different concentrations of ascorbate ranging from 0 to 0.4% (w/v) ascorbate, added into drinking water, until the age of four months. We performed label-free Liquid Chromatography-Tandem Mass Spectrometry (LC-MS/MS) global quantitative proteomic profiling to identify and quantify proteins in microsomal enriched liver extracts (MEE) from our different ascorbate treated cohorts of Gulo-/- females and males. We found that the MS intensities of several proteins in the mitochondrial complex III of the electron transport chain correlated positively with liver ascorbate concentrations in both Gulo-/- females and males. Consequently, the mitochondrial complex III activity in Gulo-/- female and male mice treated with suboptimal hepatic concentrations of ascorbate was significantly lower than Gulo-/- mice treated with optimal ascorbate concentration. Finally, proteins involved in complement activation correlated negatively with liver ascorbate concentrations in both Gulo-/- males and females.

Developmental gene expression profiles of the human pathogen Schistosoma japonicum

Project description:The life cycle of schistosomes is complex, being characterised by a series of distinct parasitic and free-living phases involving an invertebrate snail host, water and a mammalian host. A custom designed oligonucleotide microarray was utilized to profile developmental gene expression in the Asian blood fluke, Schistosoma japonicum during these parasitic and free-living transitions. Total RNAs were isolated from lung schistosomula, juvenile females and males (paired but little or no egg production), adult males and females (paired with full scale egg production), eggs, miracidia, sporocysts and cercariae. We focused on the three distinct environmental phases of the lifecycle - aquatic/snail (eggs, miracidia, sporocysts, cercariae), juvenile (lung schistosomula and paired pre-egg laying adults) and adult (paired males and females, both examined separately) stages.

Genome wide gene expression analysis BQ.Ncf1.m1J mutated and BQ wild type mice

Project description:Whole blood and spleen tissue was collected from naïve untreated mice. We compared the gene expression profiles of naïve autoimmunity prone BQ.Ncf1.m1J mutated mice and wild type BQ mice. The samples from specific pathogen-free animal facility were collected in two sample sets (marked I and II respectively) and the germ-free facility samples (marked GF / germ free) formed the third analyzed batch. The data revealed a pronounced type I interferon signature in the Ncf1.m1J mutated mice housed in both conditions.

Androgen receptor blockade promotes response to BRAF/MEK-targeted therapy

Project description:We studied a group of melanoma patients treated with neoadjuvant BRAF/MEK-targeted therapy (NCT02231775, n=51), and observed significantly higher rates of major pathologic response (MPR= <10% viable tumor at resection) and improved recurrence-free survival (RFS) in females versus males (MPR-66% versus 14%, p=0.001; RFS-64% versus 32% at 2 years, p=0.021). Findings were validated in a several additional cohorts 2-4 patients with unresectable metastatic melanoma treated with BRAF and/or MEK-targeted therapy (n=664 patients in total), demonstrating improved progression-free survival (PFS) and overall survival (OS) in females versus males in several of these studies. Studies in pre-clinical models demonstrated significantly impaired anti-tumor activity in male versus female BRAF/MEK-treated mice (p=0.006), with significantly higher expression of androgen receptor (AR) in tumors of male and female BRAF/MEK-treated mice versus control (p=0.0006 and 0.0025). Pharmacologic inhibition of AR signaling improved responses to BRAF/MEK-targeted therapy in male and female mice (p=0.018 and p=0.003), whereas induction of AR signaling (via testosterone administration) was associated with significantly impaired response to BRAF/MEK-targeted therapy in males and females (p=0.021 and p<0.0001).
