Project description:Transcriptional profiling of P. gingivalis cells comparing cells grown in anaerobic conditions to cells grown in anaerobic conditions and exposed to nitrite and to GSNO. Two-condition experiment, anaerobically vs. anaerobically exposed to nitrosative stress cells. Biological replicates: 1 independently grown and harvested bacterial cells for each nitrosative exposure tested. Four technical replicates for each experiment.

Project description:A strain of UPEC CFT073 lacking the three known NO detoxifiaction mechanisms, Hmp, FlRd and Nrf is used to study the global effect of NO on the pathogen Cultures were anaerobically grown in triplicates under two conditions: Without nitrosative stress or with nitrosative stress, induced by anaerobic growth on nitrate. RNA was extracted from the samples and following rRNA depletion, samples were analyzed by RNA-seq for genome wide transcriptional differences.

Project description:DNA microarray analysis was employed to investigate the transcriptome response to nitrosative stress in a non-denitrifying facultative photosynthetic bacterium Rhodobacter sphaeroides 2.4.1. We focused on the role played by a nitric oxide-response transcriptional regulator NnrR in the response. The transcriptome profiles of R. sphaeroides 2.4.1 and its nnrR mutant before and after exposure to nitrosating agents S-nitrosoglutathione (GSNO) or sodium nitroprusside (SNP) under semiaerobic conditions were analyzed.

Role of HcpR in nitrosative stress protection

Project description:HcpR has been shown to be required for growth of Porphyromonas gingivalis in the presence of nitrite (Infect Immun. 2012 Sep;80(9):3319-31. doi: 10.1128/IAI.00561-12. Epub 2012 Jul 9). It also has been shown to regulate the expression of hcp. To define the entire regulon of HcpR RNAseq examination of the wild type and mutant (deficient in HcpR) strains have been used to define the regulon in response to nitrite exposure in bacteria grown in the presence and absence of nitrite. Four biological replicates were prepared for both, wild type and mutant strains grown in the presence and absence of nitrite.

Project description:While in transit within and between hosts, uropathogenic E. coli (UPEC) encounter multiple stresses, including substantial levels of nitric oxide and reactive nitrogen intermediates. Strains of UPEC become conditioned to high concentrations of acidified sodium nitrite (ASN), a model system used to generate nitrosative stress. We used microarrays to define the expression profile of UPEC that have been conditioned for growth in ASN.
