Project description:Peripheral blood samples were taken from a total of 48 individuals (24 AD patients and 24 controls) and subjected to access DNA CpG methylation state using Infinium HumanMethylation450 BeadChips. This array consists of probes to determine the site-specific methylation status of more than 450K CpG loci across the human genome. Approximately 500 ng of bisulfate-converted DNA (BCD) was used per BeadChip for hybridization in accordance with the protocol and instructions of Infinium HD methylation. After completing the whole-genome amplification step and then controlled enzymatic end-point fragmentation, BCD samples were precipitated and re-suspended for hybridization for 16 h at 48C. During this process, the DNA molecules are annealed to two bead types with methylated loci (C)-specific and unmethylated loci (T)-specific oligomers. Afterward, the non-hybridized or mis-hybridized DNA was washed away, followed by single-base extension using 2,4-dinitrophrnol- and biotin-labeled ddNTPs and the annealed BCD as templates.

Project description:Genome wide DNA methylation profiling of obstructive sleep apnea (OSA) patients and healthy subjects. The Illumina Infinium 27k Human DNA methylation Beadchip v1.2 was used to obtain DNA methylation profiles across approximately 27,000 CpGs in peripheral blood mononuclear cell samples. Samples included 8 normal subjects and 16 patients with obstructive sleep apnea syndrome.

Project description:The miRNA microarray expression data used in this study was derived using the commercial BeadArray-based miRNA microarray platform (Illumina). The data set contained 211 RNA samples from peripheral blood mononuclear cells (PBMC) as described previously, including 128 schizophrenia subjects and 83 non-psychiatric controls. As each miRNA array matrix held 96 sample arrays, the samples were distributed into four array matrices.

Transcription profiling of human peripheral blood mononuclear cells from radiation-exposed workers versus control subjects

Project description:We used peripheral blood mononuclear cells to study the gene expression profiles of radiation-exposed workers versus control subjects. Human PBMC were isolated from 56 healthy donors who were working in a local Hospital (Bologna, Italy): 28 individuals were x and g radiation-exposed workers, recruited from the Nuclear Medicine and Cardiovascular Units, while the control group consisted of 28 unexposed subjects, working at the same health care facility, matched for sex, age and smoking habits. The mean dosimeter reading, expressed as accumulated doses, during the entire time of employment (range: 1-32 years), was 19.49±37.59 mSv (mean ± SD). All procedures related to the collection of blood samples were performed with an informed consent and according to the protocol approved by the Institutional Review Board.
