Microtubules and actin filaments are the two main cytoskeleton networks supporting intracellular architecture and cell polarity. The centrosome nucleates and anchors microtubules and is therefore considered to be the main microtubule-organizing centre. However, recurring, yet unexplained, observations have pointed towards a connection between the centrosome and actin filaments. Project description:Histone replacement by transition proteins (TPs) and protamines (Prms) constitutes an essential step for the successful production of functional male gametes, yet nothing is known on the underlying functional interplay between histones, TPs and Prms. Here, by studying spermatogenesis in the absence of H2A.L.2, a spermatid-specific histone variant, we identify a new step, which is essential for the transformation of nucleosomes to nucleoprotamines. H2A.L.2, synthesized at the same time as TPs, enables the invasion of nucleosomes by TPs, which themselves drive the recruitment and processing of Prms. H2A.L.2 incorporation initiates a series of successive transitional states that ultimately shift to a more stable state characterized by nucleoprotamine assembly and histone-TP displacement, leading to the generation of mature spermatozoa. Hence, the current view of histone-to-nucleoprotein transition should be revisited and include a new step with H2A.L.2 assembly prior to the action of TPs and Prms.

Mass spectrometry-based proteomic characterization of plastid-encoded RNA polymerase purified from Sinapis alba.

Project description:RNA polymerases (RNAPs) transcribe DNA into RNA and are found in all living organisms with several degrees of complexity from single polypeptide chain to multimeric enzymes. In chloroplast of angiosperms, two RNAPs are involved in plastid gene transcription: the nuclear-encoded RNA polymerase (NEP) and the plastid-encoded RNA polymerase (PEP). The PEP is a prokaryotic-type multimeric RNAP found in different states depending on light stimuli and cell identity. One of these active states requires the assembly of nuclear-encoded proteins named PEP-Associated Proteins (PAPs) with the catalytic core, triggering the timely transcription of photosynthesis-associated plastid genes in cells acquiring their photosynthetic apparatus. A purification procedure was used to enrich native PEP from Sinapis alba chloroplasts. Mass spectrometry-based proteomic analysis of the obtained fraction identified the expected subunits, i.e. the core components and the PAPs as the most abundant proteins.

Extra centrosomes induce PIDD1-mediated inflammation and Immunosurveillance

Project description:Unscheduled increases in cellular ploidy underlie defects in tissue function, premature aging, and malignancy. A concomitant event to polyploidization is the amplification of centrosomes, the main microtubule organization center in animal cells. Supernumerary centrosomes are frequent in tumors, correlating with higher aggressiveness and poor prognosis. However, extra centrosomes also exert an onco-protective effect by activating the p53 network and eliciting cell cycle arrest. Here, we report that extra centrosomes, arising during unscheduled polyploidization or aberrant centriole biogenesis, induce activation of NF-κB signaling and sterile inflammation. This signaling requires the NEMO-PIDDOsome, a multi-protein complex composed of PIDD1, RIPK1, and NEMO/IKKγ. Remarkably, the presence of supernumerary centrosomes suffices to induce a paracrine chemokine and cytokine profile, able to polarize macrophages into a pro-inflammatory phenotype. Furthermore, extra centrosomes, accumulating after cytokinesis failure, increase immunogenicity of cancer cells and render them more susceptible to NK-cell attack. Hence, extra centrosomes, the PIDDosome acts as a dual effector, able to engage not only the p53 network for cell cycle control but also NF-κB signaling to instruct innate immunity.

Gene expression in mitotic tissues of Drosophila larvae without centrosomes or too many centrosomes

Project description:Centrosome defects are a common feature of many cancers. Surprisingly, flies can proceed through the majority of development without centrosomes or with amplified centrosomes in most of their cells. It is unclear whether this is because centrosome defects do not cause many problems in Drosophila cells, or because they can adapt to cope with any problems that arise. Indeed, centrosome loss and centrosome amplification predispose fly brain cells to form tumours. Here we assess how centrosome loss or centrosome amplification perturbs cell physiology by profiling the global transcriptome of Drosophila larval brains and imaginal discs that either lack centrosomes or have too many centrosomes. Mitotic tissues (brains and imaginal discs) were dissected from 3rd instar Drosophila larvae of mutants lacking centrosomes (DSas-4 and DSas-6), a strain with too many centrosomes (SakOE) and two different wild type strains (w67 and OregonR). We extracted RNA from three biological replicates per strain and used it for hybridisation to Affymetrix Drosophila Genome 2.0 arrays. Per biological sample, material dissected from ten larvae was pooled. Gene expression of the mutant strains was compared to both wild type controls.

Proteomic profiling of centrosomes across multiple cell and tissue types by a new affinity capture method

Project description:Centrosomes are small cytoplasmic organelles that play fundamental roles in a range of cellular processes. Nearly all vertebrate cell types contain centrosomes, but whether centrosome composition and function differ between cell types, tissues and pathologies remains an outstanding question. Until now, probing centrosome composition has relied on proteomic profiling of centrosomes obtained by consecutive sucrose density gradient centrifugations, requiring up to one billion cells, thus rendering studies with multiple conditions cumbersome. Here we describe centrosome affinity capture (CAPture)-mass spectrometry (MS), a method that achieves high-coverage proteomes from 20-30 million cells. Utilising a biotin-labelled peptide derived from the CCDC61 protein, CAPture isolates intact centrosomes in a manner dependent on Ninein, a conserved centrosome component. CAPture-MS performs well across several untransformed and primary cell lines, thereby enabling comparisons of high-resolution centrosome proteomes. Using distal appendage proteins as an example, we demonstrate the utility of CAPture-MS for dissecting hierarchical interactions within the centrosome. Overall CAPture-MS represents a powerful tool to unveil temporal, regulatory, cell type- and tissue-specific changes in centrosome proteomes in health and disease.

Ubi complex characterization

Project description:Ubiquinone (UQ) is a polyprenylated lipid that is conserved from bacteria to humans and is crucial to cellular respiration. How the cell orchestrates the efficient synthesis of UQ, which involves the modification of extremely hydrophobic substrates by multiple sequential enzymes, remains an unresolved issue. Here, we demonstrate that seven Ubi proteins form the Ubi complex, a stable metabolon that catalyzes the last six reactions of the UQ biosynthetic pathway in Escherichia coli. In addition to five Ubi enzymes, the 1 MDa Ubi complex contains the newly identified UbiJ and UbiK proteins that we show to be involved in the binding of UQ intermediates and the structuration of the multiprotein complex. We also develop an in vitro assay and show that UQ biosynthesis occurs in the cytoplasmic fraction of E. coli, from which we purify the Ubi complex. Collectively, our work documents a rare case of stable metabolon and revisits the current thinking that the hydrophobic intermediates of the UQ pathway are metabolized in association with the membrane.

Proteomic profiling of centrosomes isolated from HAP1 cells

Project description:Centrosomes are cytoplasmic membraneless organelles that comprise a pair of centrioles, and a pericentriolar material. We developed a new centrosome affinity capture method termed CAPture that achieves high-coverage proteomes from 20-30 million cells. Here we present CAPture-MS data from HAP1 cells (WT and two CEP83-KO clones) demonstrating the power and sensitivity of CAPture-MS to detect centrosomal distal appendages hierarchical interactions.
