Proteome Analysis of stromal cells embedded in nanocomposite hydrogels stimulated with ultrasound

ABSTRACT: The regeneration of cartilage is a crucial aspect in the treatment of osteoarthritis. In recent years, cartilage regeneration has been pursued through several bioengineering strategies and using different stem cell types, such as adipose tissue-derived mesenchymal stromal cells (ASCs), however with a rather inefficient expression of key chondrogenic markers. Here, we analyzed the proteome of ASCs embedded in a nanocomposite hydrogel stimulated with ultrasound waves, focusing on pathways related to cartilage regeneration.

The use of piezoelectric nanomaterials combined with ultrasound stimulation is emerging as a promising approach for wirelessly triggering the regeneration of different tissue types. However, it has never been explored for boosting chondrogenesis. Furthermore, the ultrasound stimulation parameters used are often not adequately controlled. In this study, we show that adipose-tissue-derived mesenchymal stromal cells embedded in a nanocomposite hydrogel containing piezoelectric barium titanate nanop  ...[more]
