Project description:In Arabidopsis, SYNTAXIN OF PLANTS 132 (SYP132) is an evolutionarily ancient SNARE that functions with syntaxins SYP121 and SYP122 at the plasma membrane. Whereas SYP121 and SYP122 mediate overlapping secretory pathways, albeit with differences in their importance in plant-environment interactions, the SNARE SYP132 is absolutely essential for plant development and survival. SYP132 promotes endocytic traffic of the plasma membrane H+-ATPase AHA1 and aquaporin PIP2;1, and it coordinates plant growth and bacterial pathogen immunity through PATHOGENESIS-RELATED1 (PR1) secretion. Yet, little else is known about SYP132 cargoes. Here, we used advanced quantitative Tandem Mass Tagging (TMT) mass spectrometry (MS) combined with immunoblot assays to track native secreted cargo proteins in the leaf apoplast from wild type, syp121/syp122 double mutant and Arabidopsis transgenics that exhibit altered SYP132 functions, SYP132-OX and SYP132Deterrent.

Project description:This study examines how histone modification confers salt stress in Arabidopsis. The floral initiator SKB1 is found to mediate the plant’s response to salt stress by altering the methylation status of histone H4R3 and of the small nuclear ribonucleoprotein (snRNP) LSM4, and thereby affecting the expression of stress-responsive genes. Total RNA was isolated with Trizol reagent (Invitrogen) from eleven11-day-old seedlings of the wild type and skb1-1 mutant without or with 200 mM NaCl for 6 hrs. 29k Arabidopsis Genome Array hybridization was carried out by CapitalBio Corporation (Beijing, China).

Project description:We identified 296 unique proteins separated by shotgun proteomics technique, 30 surface exposed proteins using biotinylation and 67 proteins by Triton X-114 detergent phase partitioning. Combination of different separation techniques with divers in silico analyses revealed 25 possibly beta-barrel outer membrane proteins and 11 proteins enclosing signal peptide sequence. Among these signal peptide containing surface proteins we detected several proteins affecting virulence, like outer membrane protein B, 60 kDa chaperonin GroEL, putative surface antigen and peptidoglycan-associated lipoprotein. The most interesting finding is a unique antigenic 44kDa uncharacterized protein, reacting with serum specimens from R. akari infected patients. This protein is characterized for the first time in present study.

Project description:The soluble N-ethylmaleimide-sensitive factor (NSF) attachment protein (SNAP) receptor (SNARE) proteins play a central role in most forms of intracellular membrane trafficking, a key process that allows for membrane and biocargo shuffling between multiple compartments within the cell and extracellular environment. The structural organization of SNARE proteins is relatively simple, with several intrinsically disordered and folded elements (e.g., SNARE motif, N-terminal domain, transmembrane region) that interact with other SNAREs, SNARE-regulating proteins and biological membranes. In this review, we discuss recent advances in the development of functional peptides that can modify SNARE-binding interfaces and modulate SNARE function. The ability of the relatively short SNARE motif to assemble spontaneously into stable coiled coil tetrahelical bundles has inspired the development of reduced SNARE-mimetic systems that use peptides for biological membrane fusion and for making large supramolecular protein complexes. We evaluate two such systems, based on peptide-nucleic acids (PNAs) and coiled coil peptides. We also review how the self-assembly of SNARE motifs can be exploited to drive on-demand assembly of complex re-engineered polypeptides.

Project description:Metal-catalyzed oxidation reactions (MCO) is one of the most applicable methods to induce protein oxidative modifications (e.g., protein oxidation, protein oxidative cleavage and crosslinking), which are essential techniques for many applications in biotechnology and redox proteomics. However, a suitable ligand is needed to assure high stability and enhance the metal catalytic efficiency in order to induce these oxidative modifications in a selective manner. Herein, we report the influence of the polyoxometalates (POMs) as inorganic ligands on the catalytic efficiency of Cu ions to induce oxidative cleavage of a protein. In the presence of ascorbic acid as (Asc), the Cu-substituted POM (Cu-POM), α2-K8P2W17O61(Cu2+.OH2), (CuWD), cleaved hen egg white lysozyme (HEWL), a protein consisting of 129 amino acids, producing only four peptide fragments. Analyzing the intact protein, in absence and presence of CuWD/Asc, and the peptide four peptide fragments via nLC-MS/MS revealed that CuWD/Asc induced oxidative modifications and cleavage at and/or near CuWD/HEWL binding sites

Project description:The Casein Kinase 1 (CK1) family of Ser/Thr protein kinases are implicated in the regulation of many cellular processes, including cell cycle, circadian rhythm, Wnt and Sonic Hedgehog signalling. Regarded as constitutively active kinases, their regulation in cells is critically important but poorly understood. We report here that members of the FAM83 family of uncharacterized proteins are central regulators of CK1 isoforms in cells. The eight members of the FAM83 family (A-H) interact and co-localize with different CK1 alpha, alpha-like, delta and epsilon isoforms. We demonstrate that the interaction with CK1 isoforms is mediated through a number of residues within a conserved domain of unknown function, termed DUF1669, which characterises the FAM83 family. CK1-binding deficient mutants of FAM83 proteins interfere with the subcellular localization of CK1 isoforms as well as FAM83 proteins themselves and their cellular functions. We propose that DUF1669 be renamed the Polypeptide Anchor of CK1 (PACK1) domain.

Project description:Sjögren's Syndrome (SS) is an autoimmune exocrinopathy characterized by the progressive damage of salivary and lacrimal glands associated with lymphocytic infiltration. It can be defined as primary SS (pSS) or associated/secondary SS (sSS) if combined with another systemic autoimmune disease. Identifying new non-invasive biomarkers for SS diagnosis remains a challenge, and alterations in saliva composition reported in patients turn this fluid into a source of potential biomarkers. To examine the overall panorama of proteins in saliva, samples of control, pSS, and sSS individuals were analyzed by gel-based LC-MS/MS.

Project description:SNARE (soluble N-ethylmaleimide-sensitive factor attachment protein receptor) proteins are a highly conserved set of membrane-associated proteins that mediate intracellular membrane fusion. Cognate SNAREs from two separate membranes zipper to facilitate membrane apposition and fusion. Though the stable post-fusion conformation of SNARE complex has been extensively studied with biochemical and biophysical means, the pathway of SNARE zippering has been elusive. In this review, we describe some recent progress in understanding the pathway of SNARE zippering. We particularly focus on the half-zippered intermediate, which is most likely to serve as the main point of regulation by the auxiliary factors.

Project description:Frizzleds (FZD) function as receptors for secreted lipoglycoproteins of the Wingless/Int-1 (WNT) family, initiating an important signal transduction network in multicellular organisms. FZDs are classified as G protein-coupled receptors (GPCRs), which control a wide variety of physiological functions. GPCRs are well known to be regulated by phosphorylation, which can lead to specification of downstream signaling or receptor desensitization. The role and underlying mechanisms of phosphorylation of FZDs remains largely unexplored. Here, we investigated the phosphorylation of human FZD6. In order to assess FZD6 phosphorylation experimentally, we employed mass spectrometry. HEK293 cells expressing FZD6-GFP were treated with either control, WNT-3A or WNT-5A containing conditioned medium for 30 min and processed for LC-MS/MS analysis. Six phosphorylated serine residues were detected in the C-terminus of FZD6 − S592, S620, S629, S648, S653 and S656.
