Project description:CsrRS is a two component regulator which is known to regulate 10-18 % of the genome of the closely related Streptococcus pyogenes. We aimed to look for transcriptional differences between pairs of naturally occurring Streptococcus equi strains where one possessed a SNP in csrRS whereas the other did not. Such SNPs have not been identified in isolates from acute disease but have been identified in persistent isolates.

Project description:Streptococcus equi subspecies equi (S. equi) is a major pathogen which cause strangles, a highly contagious respiratory infection, in horses and other equines. In this study, we purified the extracellular vesicles (EVs) of S. equi ATCC 39506 and evaluated them as vaccine candidates against S. equi infections in mice. Through immunization in an animal model and immunoprecipitation-mass spectrometry, we evaluated EV as vaccine candidates against S. equi infections and identified novel immunogenic proteins.

Project description:Adaptation of group A Streptococcus (GAS) to its human host is mediated by two component systems that transduce external stimuli to regulate bacterial physiology. Among such systems, CsrRS (also known as CovRS) is the most extensively characterized for its role in regulating ~10% of the GAS genome including several virulence genes. Here we show that extracellular magnesium and the human antimicrobial peptide LL-37 have opposing effects on phosphorylation of the response regulator CsrR by the receptor kinase CsrS. Genetic inactivation of CsrS phosphatase or kinase activity, respectively, had similar, but more pronounced effects on CsrR phosphorylation compared to growth in magnesium or LL-37. These changes in CsrR phosphorylation were correlated with repression or activation of CsrR-regulated genes as assessed by NanoString analysis. Chromatin immunoprecipitation and DNA sequencing (ChIP-seq) revealed CsrR binding at CsrRS-regulated promoters and lower affinity binding at many other locations on the GAS chromosome. Because ChIP-seq did not detect CsrR binding at promoters associated with some CsrR-regulated genes, we investigated whether these genes might be controlled indirectly by intermediate regulators whose expression is modulated by CsrR. Transcriptional profiling of mutant strains deficient in expression of either of two previously uncharacterized transcriptional regulators in the CsrR regulon indicated that one or both proteins participated in regulation of 22 of the 42 CsrR-regulated promoters for which no CsrR binding was detected by ChIP-seq. Taken together, these results illuminate CsrRS-mediated regulation of GAS gene expression through modulation of CsrR phosphorylation, CsrR binding to regulated promoters, and control of intermediate transcriptional regulators.

Project description:Streptococcus equi subspecies equi (S. equi) is a major pathogen which cause strangles, a highly contagious respiratory infection, in horses and other equines.In this study, we discovered potential vaccine candidates using comprehensive proteomics and reverse vaccinology. As the initial step, we divided proteome of S. equi ATCC 39506 into whole cell lysate, secretory proteome, membrane proteome and extracellular vesicle and then, comparative proteomic analysis was performed to characterize the functional features of the proteome. Especially, extracellular vesicle of S. equi was evaluated at the first time. Total 114 potential vaccine candidates (PVCs) were selected using reverse vaccinology and knowledge based annotations. Comprehensive proteomic analysis confirmed that 60 PVCs were identified in S. equi ATCC 39506. Particularly, 32 PVCs were enriched in the EV proteome, suggesting that this cellular fraction may serve as vaccine.

Project description:Streptococcus equi subsp. equi (SEE) is a host-restricted bacterium that causes the common infectious upper respiratory disease known as strangles in horses. Perpetuation of SEE infection appears attributable to inapparent carrier horses because it does not persist long-term in the environment, infect other host mammals or vectors, and result in short-lived immunity. Whether pathogen factors enable SEE to remain in horses without causing clinical signs remains poorly understood. Thus, our objective was to use next-generation sequencing technologies to characterize the transcriptome of isolates of SEE from horses with acute clinical strangles and inapparent carrier horses to assess pathogen-associated changes that might reflect adaptions of SEE to the host contributing to inapparent carriage. RNA sequencing of SEE isolates from Pennsylvania demonstrated no genes that were differentially expressed between acute clinical and inapparent carrier isolates of SEE.

Project description:CsrRS is a two-component regulatory system that controls expression of multiple genes and affects virulence in group B Streptococcus (GBS), a major cause of neonatal sepsis and of invasive infection in chronically ill adults. To better understand the CsrRS regulon and its importance in GBS pathogenicity, we used GBS genomic microarrays to compare global gene expression in GBS strains 2603V/R and 515 with that in CsrR or CsrS isogenic mutants. These results and comparison with data reported previously for GBS strain NEM316 demonstrated a conserved CsrRS regulon of approximately 30 genes including those encoding beta-hemolysin, CAMP factor, and C5a-peptidase, as well as a variable repertoire of genes regulated in one or two of the three strains. Inactivation of CsrS had a similar but weaker effect at most regulated loci, but, in some cases, resulted in more pronounced or divergent regulation compared to CsrR. We conclude that CsrRS regulates a core group of genes including important virulence factors in diverse strains of GBS, but also displays marked variability in the repertoire of regulated genes and in the relative effects of CsrS signaling on CsrR-mediated gene regulation. Such variation is likely to play an important role in strain-specific adaptation of GBS to particular host environments and pathogenic potential in susceptible hosts.
