Project description:Pathogens secrete effector proteins into host cells in order to suppress host immunity and promote pathogen virulence, although many features at the molecular interface of host-pathogen interactions remain to be characterized. In a yeast two-hybrid assay, we found that the Pseudomonas syringae effector HopZ1a interacts with the Arabidopsis transcriptional regulator Abscisic Acid Repressor 1 (ABR1). Further analysis revealed that ABR1 interacts with multiple P. syringae effectors, suggesting that it may be targeted as a susceptibility hub. Indeed, loss-of-function abr1 mutants exhibit reduced susceptibility to a number of P. syringae strains. The ABR1 protein comprises a conserved APETALA2 (AP2) domain flanked by long regions of predicted structural disorder. We verified the DNA-binding activity of the AP2 domain and demonstrated that the disordered domains act redundantly to enhance DNA binding and to facilitate transcriptional activation by ABR1. Finally, we compared gene expression profiles from wild-type and abr1 plants following inoculation with P. syringae, which suggested that the reduced susceptibility of abr1 mutants is due to the loss of a virulence target rather than an enhanced immune response. These data highlight ABR1 as a functionally important component at the host-pathogen interface.

Project description:Phytopathogens use secreted effector proteins to suppress host immunity and promote pathogen virulence, and there is increasing evidence that the host-pathogen interactome comprises a complex network. In an effort to identify novel interactors of the Pseudomonas syringae effector HopZ1a, we performed a yeast two-hybrid screen that identified a previously uncharacterized Arabidopsis protein that we designate HopZ1a Interactor 1 (ZIN1). Additional analyses in yeast and in planta revealed that ZIN1 also interacts with several other P. syringae effectors. We show that an Arabidopsis loss-of-function zin1 mutant is less susceptible to infection by certain strains of P. syringae, while overexpression of ZIN1 results in enhanced susceptibility. Functionally, ZIN1 exhibits topoisomerase-like activity in vitro. Transcriptional profiling of wild-type and zin1 Arabidopsis plants inoculated with P. syringae indicated that ZIN1 regulates a wide range of pathogen-responsive biological processes, although the list of genes more highly expressed in zin1 versus wild-type plants was particularly enriched for ribosomal protein genes. Altogether, these data illuminate ZIN1 as a potential susceptibility hub that interacts with multiple effectors to influence the outcome of plant-microbe interactions.

The manner in which these signals regulate T3SS activity is still unclear. Project description:Pseudomonas syringae pv. aptata is a member of the sugar beet pathobiome and the causative agent of leaf spot disease. Like many pathogenic bacteria, P. syringae relies on the secretion of toxins, which manipulate host-pathogen interactions, to establish and maintain an infection. This study analyzes the secretome of six pathogenic P. syringae pv. aptata strains with different defined virulence capacities in order to identify common and strain-specific features, and correlate the secretome with disease outcome. All strains show a high type III secretion system (T3SS) and type VI secretion system (T6SS) activity under apoplast-like conditions mimicking the infection. Surprisingly, we found that low pathogenic strains show a higher secretion of most T3SS substrates (19 of the 23 detected effectors and accessory harpin proteins), whereas a distinct subgroup of four effectors is exclusively secreted in medium and high pathogenic strains. Similarly, we detected two T6SS secretion patterns: while one set of proteins was highly secreted in all strains, another subset consisting of known T6SS substrates and previously uncharacterized proteins with a highly similar secretion pattern was exclusively secreted in medium and high virulence strains. Taken together, our data shows that P. syringae pathogenicity is correlated with the repertoire and fine-tuning of effector secretion and indicates distinct strategies for establishing virulence of P. syringae pv. aptata in plants.

Project description:Pseudomonas syringae, a Gram-negative plant pathogen, infects more than 50 crops with its type III secretion system (T3SS) and causes severe economic losses around the world. Although the mechanisms of virulence-associated regulators of P. syringae T3SS have been studied for decades, the crosstalk and network underlying these regulators are still elusive. Previously, we have individually studied a group of T3SS regulators, including AefR, HrpS, and RhpRS. In the present study, we found 4 new T3SS regulator genes (envZ, ompR, tsiS and phoQ) via transposon-mediated mutagenesis. Two-component systems EnvZ and TsiS natively regulate T3SS. In order to uncover the crosstalk between 16 virulence-associated regulators, (including AefR, AlgU, CvsR, GacA, HrpL, HrpR, HrpS, MgrA, OmpR, PhoP, PilR, PsrA, RhpR, RpoN, TsiR and Vfr) in P. syringae, we mapped an intricate network named PSVnet (Pseudomonas syringae Virulence Regulatory Network) by combining differentially expression genes in RNA-seq and binding loci in ChIP-seq of all regulators.

Project description:The plant pathogen Pseudomonas syringae secretes multiple effectors to suppress plant defenses. Some effectors are recognized by plants while others can function to suppress the resulting immune responses. HopZ3 of Psy B728a is an aminotransferase targeting multiple components of both plant immune complexes and Psy effectors, leading to the activation of these complexes. In Arabidopsis, HopZ3 acetylates the host RPM1 complex and Psy effectors AvrRpm1 and AvrB3 s. In P. syringae resistant tomato, the main immune complex includes PRF and PTO. We found that HopZ3 acts as a virulence factor on tomato by suppressing defenses triggered by Psy AvrPto1. HopZ3 interacts with members of the PTO complex and acetylates PTO, AvrPto1, SlRIPK and SlRIN4s in vitro and in planta. Mapping HopZ3-dependent acetylation sites in planta revealed that AvrPto1 is modified on histidine, serine and threonine residues. The most highly acetylated AvrPto1 residues are T91 and S94 in a loop essential for interaction with PTO. HopZ3 also acetylates, in planta, T204 and T199 in the PTO activation loop which is important for kinase activity and recognition of AvrPto1. In agreement with these findings, we showed that acetylation of either AvrPto1 or PTO by HopZ3 reduces their binding in vitro. Mutation of acetylation sites T91A or H125A/H135A in AvrPto1 does not affect PTO binding in vitro but decreases AvrPto1 recognition in tomato in the absence of HopZ3 and increases bacterial growth. We also found decreased in planta phosphorylation in the presence of HopZ3 for some residues in AvrPto1 and PTO involved in immune signaling. Moreover, acetylation of SlRIPK by HopZ3 reduces its activity as well as phosphorylation of HopZ3 and SlRIN4s. Psy modulation of both its own avirulence effectors and their plant targets leads to decreased defenses and increased susceptibility to infection.
