Human defensin fragments to fight ESKAPE pathogens

ABSTRACT: In this project, we developed a novel antimicrobial peptide with antibacterial and antibiofilm properties. This peptide was tested for its bactericidal activity against intestinal pathogens. We could show that the human defensin fragment could reduced acute intestinal Salmonella and established Citrobacter infection, with minimal-side-effect on the gut microbiota. Human defensins are therefore promising targets to develop new treatment options.

Neutrophil-specific defensin receptors prevent skin dysbiosis and bacterial infection

Project description:Healthy skin maintains a diverse microbiome and a potent immune system to fight off infections. In this study, we discovered that epithelium-derived antimicrobial peptides defensins activate orphan GPCRs Mrgpra2a/b on neutrophils. This signaling axis is required for effective neutrophil-mediated skin immunity and microbiome homeostasis. We generated two mutant mouse lines, Defensin conditional knockout (Def cKO, K14-cre; Def fl/fl) in which the entire Def gene cluster is conditionally deleted from keratinocytes, and Mrgpra2 double knockout (Mrgpra2 dKO). We used high-throughput RNA sequencing to evaluate the whole transcriptomes of WT and mutant animals' skin under naive condition and 24 hours post-S. aureus infection. Disruption of defensin-Mrgpra2 signaling caused reduction of a network of pro-inflammatory and antibacterial gene expression. This study demonstrates the importance of epithelial-neutrophil signaling via the defensin-Mrgpra2 axis in maintaining healthy skin ecology and promoting antibacterial host defense.

Project description:The expression of plant defensins in Saccharomyces cerevisiae has been shown to increase tolerance to Zn of yeast (Mirouze et al., 2006; Shahzad et al., 2013). The present transcriptomic approach has been conducted to uncover the mechanism underlying this phenotype. WT yeast was compared to yeast expressing the AhPFF1.1b plant defensin when grown in both control medium and medium supplemented with 20 mM ZnSO4. 4 conditions with 2 biological replicates compared: control yeast (BY4741) grown on control medium, control yeast grown in the presence of 20 mM ZnSO4, yeast expressing the AhPDF1.1b defensin grown in control medium, yeast expressing the AhPDF1.1b defensin grown in the presence of 20 mM ZnSO4

Nutrient sensing by the intestinal epithelium orchestrates mucosal antimicrobial defense via translational control of Hes1

Project description:Metabolic programs and host defense are highly integrated to ensure proper immune responses during stress. Central to these responses, mTOR regulates immune functions by sensing and integrating environmental cues, yet how these systems are coordinated at the intestinal surface remains undefined. We show that the antimicrobial peptide α-defensin is functionally sustained during nutrient deprivation due to regulation of defensin-processing enzyme MMP7 by microbiota- and host-derived factors. Unlike other antimicrobial peptides, the MMP7-α-defensin axis remains active during nutrient fluctuations, providing essential protection against enteric pathogens. Sustained Mmp7 expression requires the microbiota and is mediated by de-repression of the transcription activator Atoh1 upon attenuation of the transcriptional repressor Hes1 in intestinal epithelial cells. Hes1 levels are regulated via mTOR and controlled translationally, constituting a metabolism-translation-transcription loop. Disrupting this loop by supplying nutrients paradoxically compromises anti-bacterial defense. Together, these results uncover a regulatory circuit that couples host nutrient status to epithelial anti-microbial immunity.

Involvement of β-defensin 130 (DEFB130) in the macrophage microbicidal mechanisms for killing Plasmodium falciparum

Project description:Understanding the molecular defense mechanism of macrophages and identifying their effector molecules against malarial parasites may provide important clues for the discovery of new therapies. To analyze the immunological responses of malarial parasite-induced macrophages, we used DNA microarray technology to examine the gene profile of differentiated macrophages phagocytizing Plasmodium falciparum-parasitized erythrocytes (iRBCs). The transcriptional gene profile of macrophages in response to iRBCs represented 168 down-regulated genes, which were mainly involved in the cellular immune response, and 216 upregulated genes, which were involved in cellular proteolysis, growth, and adhesion. Importantly, the specific upregulation of β-defensin 130 (DEFB130) in these macrophages suggested a possible role for DEFB130 in malarial parasite elimination. Strikingly, differentiated macrophages phagocytizing iRBCs exhibited an increase in intracellular DEFB130 levels and DEFB130 appeared to accumulate at the site of iRBC engulfment. Furthermore, DEFB130 synthetic peptide exhibited a modest toxic effect on P. falciparum in vitro and P. yoelii in vivo, unlike scrambled DEFB130 peptide, which showed no antiplasmodial activity. Our data broaden our knowledge of the immunological response of macrophages to iRBCs and shed light on a new target for therapeutic intervention.

The role of β-defensin 103 (DEFB103) copy number variation in bull fertility

Project description:Pregnancy rates for elite bulls used in artificial insemination (AI) can vary significantly and therefore the identification of molecular markers for bull fertility and targets to improve bull selection is important. β-defensins are peptides with diverse regulatory roles in sperm function across multiple species. To explore the functional impact of DEFB103 CNV on the uterine response in vivo, 18 heifers were inseminated with sperm from bulls categorized by low, intermediate, and high CN levels. Transcriptomic analysis of uterine tissue collected 12 hours after insemination revealed significant differential expression of 58 genes (FDR<0.1) related to sperm migration, immune signaling, and chemotaxis. These findings highlight the significant role of DEFB103 CN in both sperm function and the uterine response to bull sperm, suggesting its potential influence on pregnancy outcomes in cattle.
