Project description:Understanding how the response of human cells respond to protein-unfolding stress changes with cellular age is important to gain a better understanding of the link between ageing and protein-folding diseases. The experiment was designed to assess the total mRNA levels in heat shocked cells from proliferating (early-passage) and senescent (late-passage) cultures; and was used, in conjunction with mass spectrometry data, to gain an insight into the systemic response to protein-unfolding stress. Here, total RNA was extracted from early- and late-passage human mesenchymal stem cells (n=5, donor matched) following 2 hours of heat shock at 42 degrees C. RNA from cells kept at 37 degrees C was used as control. The protein coding RNAs were then sequenced with Illumina HiSeq technology.

Project description:The abf1-1 mutant and wild type cells were grown at 30 degrees C in YPAD until OD600 reached 1.0-1.2. The same volume of 42 degrees C preheated YPAD was added for each flask. RNAseq of dauer-forming C. elegans L2d in daf-2 and daf-2;daf-42

Project description:Under adverse environmental conditions, nematodes arrest into dauer, an alternative developmental stage for diapause. Dauers endure unfavorable environment and interact with host animals to access favorable environments, thus playing a critical role in the survival of both free-living and parasitic nematodes. Here, we discovered that in Caenorhabditis elegans, daf-42 is essential for development into dauer stage, as the null mutant shows lethal phenotype during dauer entry. To examine the transcriptional changes accompanied by the absence of DAF-42 during dauer entry, we performed RNA-sequencing on daf-2 and daf-2; daf-42 worms at 52 hours after egg laying (HAE) and 60 HAE, the time at which 0% and about 40% of daf-2; daf-42 mutants form dead dauer at 25 degrees Celcius, respectively. daf-2 control worms develop into dauer stage after 60 HAE, and half of its population develop into dauer by 72 HAE, when raised at 25 degrees Celcius.

Haploidentical vs matched unrelated donors for patients with ALL: donor age matters more than donor type.

Project description:Haploidentical hematopoietic cell transplantation (HCT) with posttransplant cyclophosphamide (PTCy) graft-versus-host-disease (GVHD) prophylaxis yields a similar overall survival (OS) to HLA-matched unrelated donor (MUD) HCT with conventional prophylaxis. Given the prognostic implications of donor age, we investigated the impact of donor age (younger [<35 years, n = 868] vs older [≥35 years, n = 418]) and donor type (haploidentical [n = 373] vs MUD [n = 913]) on OS in adult patients with acute lymphoblastic leukemia (ALL). Older donor age was independently associated with significantly poor OS, whereas donor type was not. Next, we directly compared the outcomes of a younger haploidentical donor (n = 187) vs an older MUD (n = 232). In this cohort, more patients in the haploidentical group had B-cell immunophenotype (89% vs 77%, respectively, P < .001), poor cytogenetics (61% vs 51%, respectively, P = .44), Philadelphia chromosome-negative (53% vs 48%, respectively, P = .38), received bone marrow graft (42% vs 16%, respectively, P < .001), and reduced-intensity conditioning (45% vs 23%, respectively, P < .001). In the multivariate analysis, the older MUD group was associated with a significantly higher risk of chronic GVHD, higher nonrelapse mortality (NRM), lower relapse, and poorer OS. Despite a higher risk of relapse, younger donor haploidentical HCT with PTCy prophylaxis may be preferred over older MUD HCT with conventional prophylaxis in patients with ALL due to lower NRM and better OS. Further analysis comparing the effect of donor age in haploidentical PTCy vs MUD PTCy is warranted.

Project description:This experiment investigates the effect of mild cold-shock (32 degrees C, 24 hours) on the translatome of human HEK293 cells. After cold-shock, cells were treated with cycloheximide to block elongation, then sucrose density gradients were used to separate post-nuclear extracts into polysome and subpolysome fractions. The pooled polysomal fraction was labelled with Cy5, the pooled subpolysomal fraction was labelled with Cy3, and both were hybridised to the same array.
