Project description:Plant hormones and small secretory peptides often function as environmental stress mediators. Some recent reports indicate that small secretory peptides, such as CLAVATA3/EMBRYO SURROUNDING REGION-RELATED (CLE), also function as mediators of environmental stimuli. CLE３ is induced in roots by defense elicitor treatment. Plants without functional CLE3 showed compromized defense gene responses in shoots when plant roots were treated with NaSA. Here, we identified specific genes downstream of CLE3 in roots and shoots with transformed Arabidopsis plants.

Project description:Phytosulfokine (PSK) is a plant peptide hormone that contributes to plant signaling and induced various of effects, including growth, senescence, stress tolerant and defense responses. PSK is a pentapeptide with two tyrosine sulfated by tyrosylprotein sulfotransferase (TPST) enzyme. The sulfated PSK can bind to its cell surface receptors, but the transcriptional readouts remain largely unknown. Here, we treated tpst mutant that are unable to produce native sulfated PSK with synthetic active PSK to capture time-series and tissue-specific readouts of PSK signaling.

Cytokinin induction of roots and shoots

Project description:Arabidopsis Col-0 seedlings grown in liquid culture to growth stage 1.0 were treated with 5 M-5M Benzyladenine and incubated for the indicated time points. Roots and shoots of the seedlings were then harvested separately by taking each individual seedling with a soft forceps at the hypocotyl, dipping it into liquid nitrogen and breaking off the root at the inner wall of a collection tube that was swimming in liquid nitrogen. The shoot part was then striped from the forceps at the edge of another collection tubes.

Insights into Transcriptional Regulatory Pathways on Systemic Communication under Flooding in Arabidopsis

Project description:We analyzed global transcriptional changes in both shoots and roots of root-flooded Arabidopsis seedlings by microarrays. We also interpreted the significance of the systemic communication between roots and shoots by functional classification of affected genes. We performed genetic analysis with an ethylene signaling mutant, ein2-5, to correlate systemic flooding responses with ethylene signaling. We identified a class of genes that were up- or downregulated in shoots, but not affected in roots, under hypoxic conditions. A comprehensive managing program of carbohydrate metabolism was observed, providing an example of how systemic communications might facilitate the survival of plants under flooding. A proportion of long-distance hypoxic regulation was altered in ein2-5.
