Project description:Identifying factors required for spermatogenesis is important for understanding mechanisms of male fertility. Inactivation of the Mgat1 or Man2a2 gene leads to a block in spermatogenesis causing infertility in male mice. In both cases multi-nucleated germ cells (MNC) are formed and there are no sperm in the epididymis. MGAT1 GlcNAc-transferase initiates complex N-glycan synthesis and MAN2A2 mannosidase generates the substrate for MGAT2 GlcNAc-transferase which transfers GlcNAc to form a biantennary complex N-glycan. Deletion of Mgat2 can leave its substrate, a hybrid N-glycan terminating in a single GlcNAc, to accumulate or the single GlcNAc may be extended to with Gal and GlcNAc to form polylactosamine. In this paper we show that conditional deletion of Mgat2 in spermatogonia via Stra8-iCre caused a block in spermatogenesis prior to the formation of round spermatids. This phenotype differs from deletion of Mgat1 by Stra8-iCre which generates MNC of round and elongated spermatids. In addition, RNA-seq analysis of germ cells prepared at 15 days after birth revealed a unique transcriptomic landscape in Mgat2[-/-] germ cells. Bioinformatic analyses predicted potential roles for ERK and AKT activities which were validated by western analyses. By contrast, Mgat1[-/-] germ cells have reduced ERK activity and unchanged AKT activity. Therefore, the defective spermatogenesis observed following removal of MGAT1 or MGAT2 must arise from the different immature N-glycans that accumulate rather than from the loss of complex N-glycans which is common to both.

Project description:The environmental exposures and lifestyle of parents can alter the development of offspring. How this environmental information is coded into heritable messages to be transmitted by gametes remains unknown, but epigenetic mechanisms have been implicated. We recently determined that disruption of histone H3 di-methylation at lysine 4 (H3K4me2) in sperm has transgenerational consequences in the development of offspring. However, little is known about when in spermatogenesis histone methylation is established and whether epimutations induced in developing sperm are permanent. We hypothesize that epigenetic modifications to histones established in spermatogonia persist through spermatogenesis and can be transmitted to offspring via the sperm. Our objective was to determine what genomic regions bearing histone H3K4me2 in spermatogonia are also present in sperm. Methods: Using transgenic mice expressing Oct4-GFP, we isolated an enriched spermatogonia population and performed ChIP-seq for H3K4me2. Results: Our analysis revealed that H3K4me2 is located throughout the genome in spermatogonia and particularly at the transcription start site region (TSS) of more than 16,000 genes. Remarkably 44% of H3K4me2 peaks present in spermatogonia are conserved in sperm. The greatest overlap between spermatogonia and sperm occurred at the TSS with 83% similarity. Finally, we assessed the expression level of genes enriched in H3K4me2 in spermatogonia and sperm. We observed that genes with the highest enrichment in H3K4me2 in sperm are expressed at higher levels in spermatogenesis and during development. Conclusion: These findings suggest that if epimutations are induced in spermatogonia they may persist in sperm and influence the health and development of offspring.

Project description:Spermatogenesis is precisely controlled at the transcriptional, posttranscriptional, and translational levels. Here we report that N6-methyladenosine (m6A), an epitranscriptomic mark regulating gene expression, plays essential roles during spermatogenesis. We present comprehensive m6A mRNA methylomes of mouse spermatogenic cells from five developmental stages: undifferentiated spermatogonia, type A1 spermatogonia, preleptotene spermatocytes, pachytene/diplotene spermatocytes, and round spermatids. Germ cell-specific inactiva- tion of the m6A RNA methyltransferase Mettl3 or Mettl14 with Vasa-Cre causes loss of m6A and depletion of SSCs. m6A depletion dysregulates translation of transcripts that are required for SSC proliferation/differentiation. Com- bined deletion of Mettl3 and Mettl14 in advanced germ cells with Stra8-GFPCre disrupts spermiogenesis, whereas mice with single deletion of either Mettl3 or Mettl14 in advanced germ cells show normal spermatogenesis. The sper- matids from double-mutant mice exhibit impaired translation of haploid-specific genes that are essential for spermio- genesis. This study highlights crucial roles of mRNA m6A modification in germline development, potentially ensuring coordinated translation at different stages of spermatogenesis.

Project description:In order to investigate the regulatory roles of AKT1S1 during mouse spermatogenesis, we analyzed gene expression changes that caused by the deletion of Akt1s1 gene in mouse male germ cells using RNA sequencing method. The transcriptome profiling analyses showed that OCT4+ spermatogonia from mice at different ages possess varied changes in gene expression due to the deletion of Akt1s1 gene, supporting the role of AKT1S1 plays in mediating AKT-mTORC1 signaling during mouse spermatogenesis.

Project description:Vertebrate spermatogonial stem cells maintain sperm production over the lifetime of an animal but fertility declines with age. While morphological studies have informed our understanding of typical spermatogenesis, the molecular and cellular mechanisms underlying the maintenance and decline of spermatogenesis are not yet understood. We used single-cell RNA sequencing to generate a developmental atlas of the aging zebrafish testis. All testes contained spermatogonia, but we observed a progressive decline in spermatogenesis that correlates with age. Testes from some older males only contained spermatogonia and a reduced population of spermatocytes. Spermatogonia in older males are transcriptionally distinct from spermatogonia in testes capable of robust spermatogenesis. Immune cells including macrophages and lymphocytes drastically increase in abundance in testes that cannot complete spermatogenesis. Our developmental atlas reveals the cellular changes as the testis ages and defines a molecular roadmap for the regulation of spermatogenesis.
