Project description:Artificially sweetened beverages containing noncaloric monosaccharides were suggested as healthier alternatives to sugar-sweetened beverages. Nevertheless, the potential detrimental effects of these noncaloric monosaccharides on blood vessel function remain inadequately understood. Presently, we have established a zebrafish model that exhibits significant excessive angiogenesis induced by D-glucose and L-glucose. Utilizing this model, we observed that glucose and noncaloric monosaccharides could induce excessive formation of blood vessels, especially intersegmental vessels (ISVs). We analyzed and compared the differentially expressed genes of control, high D-glucose-treated and high L-glucose-treated embryos. The results revealed that 1259 and 1074 genes were up-regulated significantly in high D-glucose and high L-glucose treated embryos, respectively, compared with control。

Excess glucose or fat differentially affects gene expression and appetite regulation during zebrafish embryogenesis

Project description:Zebrafish embryos were exposed to 3% glucose (24-72 hpf) and injected 2 nL of BODIPY-FL C12:0 dissolved in canola oil directly into the yolk sac at 24 hpf. At 72 hpf embryos were collected for analysis. Quantitative PCR analysis of glucose exposed embryos demonstrated that glucose exposure activates the insulin pathway while FFA/TAG injection activates the lipolysis and beta-oxidation pathways. At 72 hpf embryos were collected and bulk RNAseq was performed on control, glucose exposed and FFA/TAGs injected embryos.

RNA-seq profiling of Tetep (resistant) and HP2216 (susceptible) rice lines at 0, 48, 72, and 96 hours post infection after M. oryzae inoculation in panicle

Project description:Purpose: The aim of this study was to compare transcriptome profiling (RNA-seq) of rice panicle tissue from resistant and suceptible lines at different time intervals infected with M.oryzae. The results were validated using Semi-quantitative PCR. Methods: Panicles from resistant and suceptible rice lines were inoculated with suspension having double distill water, Tween-20 without spores for 0 hour and same suspension with spores was used to inoculate for 48, 72 and 96 hour samples. After collection, samples were frozen on liquid nitrogen, and RNA was extracted using Spectrum™ Plant Total RNA Kit (Sigma). RNA libraries were prepared for sequencing using standard Illumina protocols. Results: Differential Expressed Genes were identified, comparing transcriptome of Control 0 hour vs 48, 72 and 96 hour post infection. Where we get 1070, 2383 and 1080 total significant genes at 48, 72, and 96 hour post infection respectively in HP2216. And, 1564, 1433 and 2263 total significant genes at 48, 72, and 96 hour post infection respectively in Tetep.
