Transcription profiling by array of Pseudomonas aeruginosa in a rhizopsphere during phosphate limitation conditions

Project description:Plants and rhizosphere microbes rely closely on each other, with plants supplying carbon to bacteria in root exudates, and bacteria mobilizing soil-bound phosphate for plant nutrition. When the phosphate supply becomes limiting for plant growth, the composition of root exudation changes, affecting rhizosphere microbial communities and microbially-mediated nutrient fluxes. To evaluate how plant phosphate deprivation affects rhizosphere bacteria, Lolium perenne seedlings were root-inoculated with Pseudomonas aeruginosa 7NR, and grown in axenic microcosms under different phosphate regimes (330 uM vs 3-6 uM phosphate). The effect of biological nutrient limitation was examined by DNA microarray studies of rhizobacterial gene expression.

Transcription profiling of Pseudomonas putida in the rhizosphere of corn

Project description:It has been performed a genome-wide analysis of gene expression of the root-colonizing bacterium Pseudomonas putida KT2440 in the rhizosphere of corn (Zea mays var. Girona. To identify reliable rhizosphere differentially expressed genes, rhizosphere populations of P. putida bacteria cells were compared with three alternative controls: i) planktonic cells growing exponentially in rich medium (LB), ii) planktonic cells in stationary phase in LB, and iii) sessile populations established in sand microcosms, under the same conditions used to grow inoculated corn plants.

DNA aptamers for detection of pyoverdine pf5

Project description:The rhizosphere is a small region surrounding plant roots that is enriched in biochemicals from root exudates and populated with fungi, nematode, and bacteria. Interaction of rhizosphere organisms with plants is mainly promoted by exudates from the roots. Root exudates contain biochemicals that come from primary and secondary metabolisms of plants. These biochemicals attract microbes, which influence plant nutrition. The rhizosphere bacteria (microbiome) are vital to plant nutrient uptake and influence biotic and abiotic stress and pathogenesis. Pseudomonas is a genus of gammaproteobacteria known for its ubiquitous presence in natural habitats and its striking ecological, metabolic, and biochemical diversity. Within the genus, members of the Pseudomonas fluorescens group are common inhabitants of soil and plant surfaces, and certain strains function in the biological control of plant disease, protecting plants from infection by soilborne and aerial plant pathogens. The soil bacterium Pseudomonas protegens Pf-5 (also known as Pseudomonas fluorescens Pf-5) is a well-characterized biological strain, which is distinguished by its prolific production of the secondary metabolite, pyoverdine. Knowledge of the distribution of P. fluorescens secretory activity around plant roots is very important for understanding the interaction between P. fluorescens and plants and can be achieved by real time tracking of pyoverdine. To achieve the capability of real-time tracking in soil, we have used a structure-switching SELEX strategy to select high affinity ssDNA aptamers with specificity for pyoverdine over other siderophores. Two DNA aptamers were isolated, and their features compared. The aptamers were applied to a nanoporous aluminum oxide biosensor and demonstrated to successfully detect PYO-Pf5. This sensor provides a future opportunity to track the locations around plant roots of P. protegens and to monitor PYO-Pf5 production and movement through the soil.

Gene Expression Patterns in Potato Roots Associated with Greater Plant Growth at Low Phosphate Concentrations in the Rhizosphere

Project description:There are both financial and environmental reasons for optimising the use of phosphorus (P) fertilisers in potato production. One strategy to effect this is to develop genotypes with greater P use efficiency (PUE), which is generally defined as yield divided by the P available to the crop. In this article we report that potato (Solanum tuberosum L.) genotypes differ in their PUE, expressed both as yield in the field and as shoot biomass when grown hydroponically. When grown hydroponically, PUE was strongly correlated with P uptake efficiency (PUpE, defined as shoot P content), but not with physiological P utilisation efficiency (PUtE, defined as shoot biomass divided by shoot P content). Gene expression in roots differed between potato genotypes, and changed with P supply. A common transcriptional response to reduced P supply was determined in roots of four potato genotypes. This response comprised genes encoding enzymes involved in increasing rhizosphere P availability, phosphate transport, P-sparing metabolism, replacement of phospholipids in cell membranes, and various transcription factors. A specific transcriptional response associated only with roots of genotypes with high yields in the absence of P-fertiliser application in the field (Maris Piper and Stirling) was also identified. Genes involved in this response encoded various transcription factors, proteins facilitating water, solute and auxin transport, and enzymes involved in polyamine metabolism and the biosynthesis of tropane alkaloids.

Arabidopsis thaliana mutant leaves treated with Pseudomonas syringae ES4326

Project description:In order to better understand the transcriptional networks triggered by pathogen inoculation, we monitored gene expression in leaves of mutant Arabidopsis plants, inoculated with Pseudomonas syringae ES4326 and wild type Col-0 plants grown in parallel. Individual leaves were injected in the morning using a needle-less syringe with 10E5 cfu cm-2 PsmES4326 (suspended in 5 mM MgSO4). For the wild type, leaves were also mock treated with 5 mM MgSO4. Leaves were harvested 24 hours later. Plants were grown in pots with BM-2 soil (Berger Peat Moss Ltd, Quebec, Canada) at a density of 9 plants per pot and kept at 22 degrees Celsius with 75% humidity and a 12 hour day length. Leaf transcriptomes of Arabidopsis thaliana colonized by representative phyllosphere commensals alone and in combination with the pathogen Pseudomonas syringae DC3000

Project description:Plants are colonized by a variety of microorganisms, the plant microbiota. In the phyllosphere, the above-ground parts of plants, bacteria are the most abundant inhabitants. Most of these microorganisms are not pathogenic and the plant responses to commensals or to pathogen infection in the presence of commensals are not well understood. We report the Arabidopsis leaf transcriptome after 3 to 4 weeks of colonization by Methylobacterium extorquens PA1 and Sphingomonas melonis Fr1, representatives of two abundant genera in the phyllosphere, compared to axenic plants. In addition, we also sequenced the transcriptome of Arabidopsis 2 and 7 days after spray-infection with a low dose of P. syringae DC3000 and in combination with the commensals.
