Exploring differentially expressed genes in the hypothalamic-pituitary-gonadal axis of estrous and anestrous primiparous sows using RNA-Seq technique

Project description:To investigate the effects of HPG axis on reproductive function and the underlying mechanisms involved, six libraries, including hypothalamic, pituitary and ovarian samples (termed A, B and C) from the sows in normal estrus and the corresponding samples (termed A1, B1 and C1) from the sows in which anestrus was caused by inactive ovaries, were constructed and sequenced to analyze the differentially expressed genes (DEGs) between different HPG axises. A total of 710 (392 up and 318 down), 707 (283 up and 424 down), and 956 (635 up and 321 down) DEGs were identiﬁed in A vs. A1, B vs. B1 and C vs. C1, respectively. Gene ontology (GO) and KEGG pathway analyses showed that the DEGs were most enrichment in neuropeptide hormone activity, ovarian steroidogenesis, FoxO signaling pathway and GnRH signaling pathway etc.

Transcriptomic analysis of the porcine anterior pituitary gland during the peri-implantation period

Project description:In pigs, the peri-implantation period is recognized as determinant of successful reproduction, and it is controlled by neuroendocrinal interactions among signals originating from the female hypothalamic-pituitary-ovarian axis, uterus, and developing embryo. The transcriptomic activity of the pituitary gland may influence reproductive processes occurring in pigs during the peri-implantation period. The aim of this study was to determine alternations in gene expression in the porcine pituitary gland during the peri-implantation period (days 15-16 of pregnancy) in comparison with that during the respective days of the estrous cycle using a microarray approach.

Project description:Adiponectin is thought to be involved in the regulation of metabolic homeostasis and reproductive processes. Adiponectin controls carbohydrate and lipid metabolism by regulating the fatty acid oxidation process, increasing insulin sensitivity and decreasing plasma triglyceride levels. Adiponectin also plays an important role in the modulation of female reproductive functions, both directly and by influencing the secretory functions of the higher branches of the hypothalamic-pituitary-gonadal axis. The main aim of this study was to determine the effect of adiponectin on global gene expression and on differentially expressed genes (DE-genes) under the influence of the studied hormone in porcine anterior pituitary (AP) cells.

The modulatory effect of visfatin (NAMPT) on the transcriptomic profile of porcine anterior pituitary cells during the peri-implantation period

Project description:Pregnancy is a unique and critical period in a female’s life during which the whole organism is mobilized and focused on ensuring the successful survival and development of the embryo. The main regulatory system responsible for the reproductive functions during both the oestrous cycle and pregnancy is the hypothalamic-pituitary-gonadal (HPG) axis. Herein we verified hypothesis assuming the modulatory effect of visfatin on the anterior pituitary transcriptome during the peri-implantation period. We analyzed samples obtained from 5 individuals (n=5). After the enzymatic tissue digestion with the use collagenase V and pancreatin cells were divided for two groups: controls [without treatment] and visfatin (100 ng/mL) treated ones. In vitro cell cultures were conducted for 24 hours.

Adrenal gland gene expression in male pigs from dams fed diets supplemented with algae or fish oil and challenged with LPS in late gestation

Project description:Maternal stress occuring during gestation can influence the functioning of the stress and immune responses in offspring. Maternal supplementation with immunomodulatory compounds such as omega-3 polyunsaturated fatty acids may reduce inflammation associated with maternal stress, promoting offspring health and growth Sows were fed diets supplemented with microalgae or fish oil from gestation day 75 and were challenged with LPS on gestation day 112. Previously, LPS challenge in the male piglets of these sows resulted in differences in fever and cortisol responses among piglets from different maternal treatments. Therefore, the adrenal gland was chosen for microarray analysis to assess the effects of the maternal treatment on the gene expression within the hypothalamic-pituitary-adrenal axis.

Project description:Primiparous sows were randomly allocated to two treatments and were separated from piglets 8h daily from Day 21 of lactation companied with daily boar exposure for oestrus detection until weaning (Day 28). Gene expression of Day 9 embryos were compared between control sows (FE; sows artificially inseminated when in heat during lactation ) and Skip-a-Heat sows (SE; sows in heat during lactation and artificially inseminated on the following oestrus cycle). Stimulating lactational oestrus then two mating strategies were applied to primiparous sows; 1)FE; sows were in heat during lactation and received artificial insemination) and Skip-a-Heat sows (SE; sows were in heat during lactation and received artificial insemination at fallowing oestrus cycle).

Proteomic profiling of anterior pituitary of heifer calves offered either a high or moderate plane of nutrition in early life

Project description:Enhanced nutrition during the early calfhood period has been shown to hasten the onset of puberty in heifer calves. The hypothalamic-pituitary-ovarian biochemical signalling axis regulates reproductive development in heifers, with the dynamics of gonadotropin pulsatility within the anterior pituitary gland, in particular, central to final sexual maturity. However, the precise molecular mechanisms regulating the influence of metabolic status on this signalling axis in heifer calves is yet to be fully elucidated. The objective of this study was to determine the effect of an enhanced plane of nutrition during early life, up to 21 weeks of age, on the anterior pituitary gland proteome of heifer calves. Heifer calves were offered either a high or moderate plane of nutrition. Pathway analysis of differentially abundant proteins highlighted an effect on both growth hormone and GnRH signalling pathways, as well as an effect towards reproductive system development and function
