Project description:We have developed a generally adaptable, novel high-throughput chromosome conformation capture assay for use in trans (V3C-seq) that allows genome-wide identification of the direct associations of a lytic virus genome with discreet regions of the cellular chromosome. Upon infection, the parvovirus Minute Virus of Mice genome associated directly with sites of cellular DNA damage. These sites also exhibited damage in uninfected cells when cycling through S-phase. As infection proceeded, new sites of DNA damage were induced, and virus subsequently also associated with these.

Project description:We have developed a generally adaptable, novel high-throughput Viral Chromosome Conformation Capture assay (V3C-seq) for use in trans that allows genome-wide identification of the direct interactions of a lytic virus genome with distinct regions of the cellular chromosome. Upon infection, we found that the parvovirus Minute Virus of Mice (MVM) genome initially associated with sites of cellular DNA damage that in mock-infected cells also exhibited DNA damage as cells progressed through S-phase. As infection proceeded, new DNA damage sites were induced, and virus subsequently also associated with these. Sites of association identified biochemically were confirmed microscopically and MVM could be targeted specifically to artificially induced sites of DNA damage. Thus, MVM established replication at cellular DNA damage sites, which provide replication and expression machinery, and as cellular DNA damage accrued, virus spread additionally to newly damaged sites to amplify infection. MVM-associated sites overlap significantly with previously identified topologically-associated domains (TADs).

The NS1 Protein of the Parvovirus MVM Aids in the Localization of the Viral Genome to Cellular Sites of DNA Damage

Project description:The autonomous parvovirus Minute Virus of Mice (MVM) localizes to cellular sites of DNA damage to establish viral replication centers, visualized by focal depositions of the essential MVM phosphoprotein NS1. However, how such foci are established remains unknown. Here, we show that MVM NS1 localized to cellular sites of DNA damage independently of its known ability to covalently bind the viral genome at its known consensus DNA binding site. NS1 that retained its capacity to bind its cognate DNA binding sequence could relocate either the viral genome, or heterologous DNA, containing those sites, to cellular sites of DNA damage. These findings suggest that another way that NS1 can facilitate virus infection is by localizing the genome to cellular sites which contain replication and expression proteins.

TP53 sequence of human glioblastoma stem cells sorted for p53-Ser15 induction and NS1 virus protein expression upon parvovirus MVMp infection

Project description:We have analyzed whether mutations in the TP53 gene and post-translational phosphorylation in the Ser-15 residue of the p53 protein (Pp53) may confer permissiveness to the prototype strain of the parvovirus Minute Virus of MIce ( MVMp) infection in human glioblastoma stem cells (GSCs) isolated from brain tumours. Infected (MVMp samples) and control uninfected (mock samples) GSCs from four patients (GS5, GS7, GS8, GS9) were sorted for the Pp53 modification (which is required for virus genome replication) and the NS1 protein (a major marker of virus cytotoxic gene expression). Cells sorted through restrictive gates for the NS1+/Pp53+ and NS1+/Pp53- phenotypes were proceeded for mRNA purification, the TP53 gene was amplified with specific primers and subjected to NGS analysis.

Project description:We have previously developed a modified iteration of a viral chromosome conformation capture (V3C-seq) assay to show that the autonomous parvovirus Minute Virus of Mice (MVM) localizes spatially with cellular sites of DNA damage to establish viral replication centers. Similar V3C-seq assays to map AAV genome localization show that both replicating and non-replicating AAV2 genomes in the absence of helper virus colocalize with cellular sites of DNA damage. The AAV non-structural protein Rep 68/78, when ectopically expressed in the absence of viral infection or during AAV2 infection in the absence of helper proteins also localizes to cellular sites of DNA damage. Strikingly however, recombinant AAV gene therapy vector genomes derived from AAV do not colocalize with AAV and Rep at cellular DDR sites.
