Project description:Flower developmental stage specific genome-wide H3K27ac loci were mapped in this study. By quantifying the enrichment of H3K27ac modification at each stage, we revealed the dynamic landscape of H3K27ac during flower development. The dynamic of H3K27ac was found to contribute to stage specific gene expression. We found that H3K27ac always locate in proximal region of transcription starting sites but hardly appear in distal intergenic region.

Chromatin profiling reveals novel proteins associated with histone-marked genomic regions

Project description:Our understanding of the functions of DNA elements is limited by the paucity of information about the spectrum of proteins that occupy these genomic regions. Here we describe an approach to identify proteins associated with genomic regions whose chromatin is marked by specific modified histones, which we term chromatin profiling. We used chromatin immunoprecipitation followed by mass spectrometry (ChIP-MS) to identify proteins associated with genomic regions marked by histone H3K27Ac, H3K4me3, H3K79me2 and H3K36me3 in mouse embryonic stem (mES) cells. We identified 385 known and 224 novel candidate proteins associated with these histone-marked genomic segments and confirmed that several of the novel candidates are indeed associated with histone-marked segments of the genome. Future study of the novel candidates, many of which have been implicated in various diseases, should lead to an improved understanding of gene control and its dysregulation in disease. Project description:Epigenetic alterations of monocytes from COVID-19 convalescent individuals and healthy controls have been analyzed by mapping trimethylated H3K4 (H3K4me3) and acetylated H3K27 (H3K27ac) genome-wide using CUT&RUN (Skene & Henikoff, 2017). Epigenetic reprogramming of myeloid cells represents a hallmark of trained innate immunity (Netea, Joosten et al., 2016). Identification of H3K27ac-marked enhancers in bovine satellite cells during differentiation

Project description:In this study we isolated satellite cells from skeletal muscle from two Angus-crossbred steers, initially cultured them in growth medium for proliferation, and then in differentiation medium to induce them to differentiate and fuse into myotubes. We used ChIP-seq to identify enhancers marked with H3K27ac, i.e., active enhancers, from these cells immediately before and two days after induction of differentiation. Nearly 20,000 and 50,000 H3K27ac-marked enhancers were consistently identified from two biological replicates of before- and during-differentiation bovine satellite cells, respectively.
