Project description:Anxiety and stress-related conditions represent a significant health burden in modern society. Unfortunately, most of the drugs currently used in the clinic modulate neurotransmission and are prone to side effects, limiting their long-term use. We used bioinformatics screening to find small molecules mimicking the transcriptional profile recently reported in hippocampi of importin α5 mutant anxiolytic mice, identifying the plant sterol β-sitosterol as a top candidate. Behavioral analyses confirmed anxiolytic properties of β-sitosterol. The present RNA-seq experiment characterizes the hippocampal transcriptome in mice 1 hr and 6 hrs after intraperitoneal injection of 100 mg/kg β-sitosterol or vehicle.

RNAseq sequencing signatures of Diffuse Large B-cell Lymphoma cells after beta-sitosterol treatment

Project description:Resistance to Rituximab is a significant challenge in treating Diffuse Large B-cell Lymphoma (DLBCL). β-Sitosterol (β-ST) is a plant sterol that has been found in a broad variety of fruits, spices, and medicinal plants. While the anti-neoplastic properties of β-Sitosterol (β-ST) are established in various solid malignancies, its impact on DLBCL is uncharted. To further investigation the mechanism of β-ST on DLBCL, we employed RNAseq transcriptome analysis to detect differential expression genes (DEGs) between β-ST treated and untreated DLBCL cells. Our findings indicate that β-ST impedes DLBCL cell proliferation in a concentration and time-dependent manner. It appears to alter sphingolipid metabolism, facilitate (ASM translocation to the plasma membrane, augment ceramide platforms through increased ceramide synthesis, and consequently precipitate apoptosis in DLBCL cells.

Transcription profiling of mouse vehicle and sAPPalpha-treated hippocampal slice cultures

Project description:Gene expression changes induced by alpha-secretase cleaved amyloid precursor protein (sAPPalpha) in organotypic hippocampal slice cultures of male, postnatal day 15 mice (C57B6/SJL). Hippocampal slice cultures were treated with phosphate buffered saline (GSM26700, GSM26701, GSM26702) or 1 nM sAPPalpha (GSM26703, GSM26704, GSM26705) for 24 h. Each sample consists of total RNA isolated from 8-12 slices from 4 mice. Data were analyzed with MAS 5.0 and scaled to 2500. sAPPalpha induces the amyloid sequestration protein transthyretin, insulin-like growth factor 2, insulin-like growth factor binding protein 2, and other genes involved in protective pathways such as apoptosis inhibition, detoxification, and retinol transport. Sub-chronic AM6545 administration enhances cognitive performance and induces hippocampal synaptic plasticity changes in male mice

Project description:Using RNA sequencing, we perform gene expression profile of five male mice treated with vehicle and five male mice sub-chronically treated with AM6545 at a dose of 1 mg/kg. To evaluate hippocampal modifications promoted by peripheral CB1R modulation, we analyze at transcript level hippocampal synaptoneurosomes fractions. This analysis revealed changes in the transcript usage of N-methyl-D-aspartate (NMDA) receptor 1 isoforms in AM6545 treated mice compared with vehicle-treated ones.
