Project description:We found that Quaking5 protein, a RNA splicing protein was downregulated in HCC. Collectively, this study uncovers the mechanisms underlying the regulating role of circZKSCAN1 in HCC cancer stem cells and reveals that the new discovered Qki5-circZKSCAN1-FMRP-CCAR1-Wnt signaling axis is of important therapeutic value in improving the treatment of HCC.
Project description:The oncogene Melanoma differentiation associated gene-9/syndecan binding protein (MDA-9/SDCBP) promotes tumorigenesis and metastasis in many cancers. However, the role of MDA-9 in regulating HCC has not been well-studied. To unravel the function of MDA-9 in HCC we generated a transgenic mouse with hepatocyte-specific overexpression of MDA-9 (Alb/MDA-9). Alb/MDA-9 mice demonstrated significantly higher incidence of N-nitrosodiethylamine/phenobarbital-induced HCC compared to WT littermates. To check what genes are regulated by MDA-9 to promote HCC, RNA-seq was performed in naïve WT and Alb/MDA-9 hepatocytes.
Project description:Here, we assessed the expression of Mac-1 on mouse HSCs during regeneration following transplantation and observed a transient increase in Mac-1 expression during the early reconstitution phase. Serial transplantation experiments demonstrated that reconstitution potential was highly enriched in the Mac-1+ portion of the HSC pool.

We evaluated innate and antigen-specific adaptive immune responses to Mycobacterium avium complex in individuals with MAC lung disease. We investigated the gene expression changes induced in PBMCs by polyclonal stimulation (anti-CD3, anti-CD28), Mav and Mtb lysates and a peptide pool (MTB300) in individuals with MAC lung disease (MACDZ), and healthy controls (IGRA+ and IGRA-).
Project description:The bovine mammary epithelial cell line Mac-T has been used to study mammary gland in vitro. The reliability of this system to study mammary gland has not been tested using genomics approaches. In the present experimetn a direct transcriptomics comparison between Mac-T cells and mammary tissue at -30 and at 60 day in milk (DIM) is performed. Data indicated that Mac-T cells and mammary tissue had a substantially different transcriptome with a larger difference between Mac-T cells and lactating mammary tissue (i.e., 60 DIM) compared to non-lactating mammary tissue (i.e., -30 DIM). In addition, data indicated that the Mac-T cells substantially differ with mammary tissue in lactation-specific functions.
