Project description:This microarray experiment is part of a study addressing the importance of glutamine metabolism in Streptococcus pneumoniae for the virulence of this bacterium. To be able to gain more insight into the phenotype of a double mutant of glnA (TIGR 4 locus tag SP0502, encoding glutamine synthetase) and glnP (SP1241, encoding a permease component of a glutmine uptake ABC transporter), the transcriptome of this mutant, the glnPA double mutant, was determined in strain S. pneumoniae D39 grown in rich GM17 medium with 0.5 mg/ml glutamine. This revealed a big change in transcriptome in the mutant, especially a lot of amino acid and peptide metabolic genes. Project description:In mouse, the adrenocortical dysplasia (acd) phenotype shows limb and body axis anomalies, as a result of p53-dependent apoptosis, and perinatal lethality. The p53 deficiency partially rescues anomalies, but not perinatal lethality, implicating the involvement of p53-independent mechanisms in the acd phenotype. Differentially expressed genes in acd mutant and double mutant embryos were identified. p53-dependent and –independent pathways contributing to acd phenotype were characterized. We used microarrays to analyze gene expression profiles of acd and double mutant embryos and to identify p53 dependent and –independent mechanisms underlying acd phenotype.
Project description:To study the function of zebrafish nuclear pores during early embryogenesis, we generated maternal zygotic double mutant of nup85;nup133 (MZnup85;nup133) using CRISPR/Cas9 and report the transcriptome-wide changes in comparison to wild-type (WT) embryos. Our analysis reveals a dramatic delay of maternal mRNA degradation and zygotic genome activation in MZnup85;nup133 embryos during maternal-to-zygotic transition.
