Project description:Semaphorin 5A (SEMA5A) is one member of semaphorin family, which was identified as an axon guidance molecule in nervous systems. In our previous studies, SEMA5A was identified as a prognostic biomarker for lung cancer in nonsmoking women and acted as a tumor suppressor gene in inhibiting the proliferation and migration of lung cancer cells. Yet, the regulatory mechanism of SEMA5A was not clear. Therefore, the purpose of this study was to explore the roles of different domains of SEMA5A on the tumor suppressive effects in lung cancer cell lines.

Project description:Proliferative stage (tachyzoite) is a critical factor in the transmission and pathogenesis of Toxoplasma gondii (T. gondii), which is capable of infecting a large variety of host cells. Proto-oncogene eukaryotic translation initiation factor (eIF-5A) is thought to function in regulation of many cellular processes including cell proliferation. Investigation the specific roles of eIF-5A involved in T. gondii is instrumental for the elucidation of tachyzoite replication. Here, eIF-5A knockdown strain was generated and analyzed by isobaric tags for relative and absolute quantification (iTraq), which suggested that proto-oncogene eIF-5A also acts as an essential gene to ensure proper protein synthesis during T. gondii invasion and replication.

Effect of 2-aryl benzimidazole (5a) on Gene Expression Profile of the Human Breast Cancer Cells

Project description:To understand the mechanism of 2-aryl benzimidazole (5a) against breast cancer cells, we used microarray gene expression analyses to discover which target or signaling pathways were significant impact by 5a in MDA-MB-453 cells. We have investigated the mRNA expression patterns before and after 5a administration of breast cancer cells, we discovered that 5a had a significant impact on the cell cycle and MEK/Erk signaling pathways, and their p-Value were 5.62 × 10-21 and 2.04 × 10-17 respectively. We also searched for genes that exhibited the greatest differences in expression of the 5a-induced MDA-MB-453 cells relative to the control cells. Several of these genes, including FOS, JUN, TNFRSF10B (DR5), CDKN1A (p21), E2F1, E2F2 and CDC20, have been reported to relate to apoptosis and cell cycle arrest.

Plexin-B3 is a functional receptor for semaphorin 5A.

Project description:Semaphorins are a large family of molecular cues implicated in neural development and in a variety of functions outside the nervous system. Semaphorin 5A (Sema5A) is a transmembrane semaphorin, containing seven thrombospondin type-1 repeats, which was recently found to control axon guidance. Here we show that plexin-B3 is a high-affinity receptor specific for Sema5A. We further demonstrate that plexin-B3 activation by Sema5A mediates functional responses in plexin-B3-expressing cells (either fibroblasts, epithelial and primary endothelial cells). In addition, Sema5A can trigger the intracellular signalling of the hepatocyte growth factor/scatter factor receptor, Met, associated in a complex with plexin-B3. We thus conclude that Sema5A is able to elicit multiple functional responses through its receptor plexin-B3.
